滑膜细胞原代体外培养体系的建立
System Construction of Human Primary Synovial Cell in Vitro
【摘要】 目的:改善体外分离、培养滑膜细胞的方法,并对其鉴定。方法:取人滑膜组织采用机械分离、复合酶消化后再将组织块塑料孔板倒贴法,差速贴壁法纯化后培养,倒置显微镜观察生长情况,HE染色,细胞活力测定,免疫组化鉴定。结果:分离培养的滑膜细胞呈梭形或柱形,核呈卵圆形位于细胞中央,生长好、纯度高。细胞活力测定大于95%,免疫组化鉴定:波形蛋白vimentin(+),CD68(-)。结论:成功分离了人滑膜细胞,方法简便易行,为进一步研究提供良好的实验模型。
【Abstract】 Objective:To improve the methods for isolation and culture of primary synovial cell and to identify it.Methods:Human synovial tissues were mechanically isolated,enzyme-digested and cells purified by invert attachment and differential attachment methods.Cell morphology and its growth characteristics were observed through invert microscope,identified by cell viability measurement,HE and immunohistochemistry staining.Results:Synovial cell isolated and cultured presented fusiform or column shape,well growed and were highly purified.Nucleus presented orbicular-ovate shape and located in the central of cell.The expressions of waveshape protein vimentin(+),CD68(-)were detected by immunohistochemistry with the cell viability of more than 95%.Conclusion:The technique of isolation and culture has the advantage of good reproducibility and provides good cell pattern for further studying.
