荧光共振能量转移法测定脂质体与细胞膜间的融合
Determination of fusion between liposomes and cell membranes by fluorescence resonance energy transfer assay
【摘要】 目的:利用荧光共振能量转移原理建立测定脂质体与细胞膜间融合能力的方法。方法:利用荧光共振能量转移原理,通过检测试样的荧光发射光信号,来评价高分子酸敏脂质体之间,高分子酸敏脂质体、红细胞及细菌之间的融合性能。结果:荧光共振能量转移法可用于测定酸性条件下酸敏脂质体与空白脂质体、红细胞膜影以及细菌之间的融合程度,本方法的可靠性得到了脂质体融合前后粒径变化的验证,方法学研究结果显示此方法的稳定性和重现性良好。结论:利用荧光共振能量转移原理,建立一种用来评价脂质体之间,脂质体与细胞膜之间融合性的方法,此方法简单、快速、有效。
【Abstract】 Objective: To establish a method for determination of the fusion between the liposomes and membrane of cell by fluorescence resonance energy transfer.Methods: To use the principle of fluorescence resonance energy transfer,the fusion between the polymer acid sensitive liposomes and red blood cells and bacteria was evaluated by detecting fluorescence emission light signals.Results: The fluorescence resonance energy transfer method can be used for determination fusion of the pH-sensitive liposomes,blank liposomes,red cell membrane and bacteria.The reliability of the fluorescence resonance energy transfer assay has been verified that liposomal particle size is changed following pH-induced fusion of acid-sensitive polymer-liposomes with non-polymer liposomes.The methodology results showed that this method was stability and good reproducibility.Conclusion:The simple and quick method to evaluate the pH-sensitivity fusion of liposome with liposomes or cell is established based on the principle of fluorescence resonance energy transfer.All results indicate that this method is simple and reliable.
