HPLC测定金银花药材中木犀草苷含量的优化

【摘要】 目的:优化金银花药材中木犀草苷的含量测定方法。方法:采用高效液相色谱法,以苯基硅烷键合硅胶为填充剂,色谱柱为Agilent ZORBAX SB-phenyl(4.6mm×250mm,5μm)柱,流动相为乙腈-0.5%醋酸溶液系统,梯度洗脱,流速1mL·min-1,检测波长350nm,柱温25℃,进样量10μL。结果:木犀草苷进样量在0.09~0.91μg范围内与峰面积呈良好的线性关系(r=0.9999);平均回收率(n=6)为103.1%,RSD为2.1%;检测限(S/N=3)为3.48ng,定量限(S/N=10)为8.70ng。结论:本法简便准确,可用于金银花药材中木犀草苷的含量测定。

【Abstract】 Objective:To develop an HPLC method for the determination of luteolin-7-O-glucoside in Flos Lonicerae Japonicae.Method:An Agilent ZORBAX SB-phenyl column(4.6 mm×250 mm,5 μm) was adopted;The mobile phase was acetonitrile-0.5% acetic acid with gradient elution mode at a flow rate of 1 mL·min-1;The detecting wavelength was 350 nm,the column was 25 ℃,and the injection volume was 10 μL.Results:The calibration curve was linear in the range of 0.09-0.91 μg of luteolin-7-O-glucoside with correlation coefficient of 0.9999;The average recovery(n=6) was 103.1% with RSD of 2.1%;The limit of detection(S/N=3) was 3.48 ng and the limit of quantification(S/N=10) was 8.70 ng.Conclusion:This method is accurate,and can be used to determine the content of luteolin-7-O-glucoside in Flos Lonicerae Japonicae.
