高效液相色谱法测定蒲公英提取物中绿原酸含量

【摘要】 目的:建立HPLC法测定蒲公英提取物中绿原酸含量。方法:采用Agilent Zorbax SB C18柱(4.6mm×250mm,5μm),流动相为乙腈-0.4%磷酸溶液(10∶90),流速1.0mL·min-1,检测波长324nm,柱温30℃。结果:绿原酸的线性范围为0.04~0.60μg(r=0.9992),平均加样回收率(n=5)为96.48%。结论:采用HPLC法测定蒲公英提取物中绿原酸含量的方法快速、简单、准确,可用于蒲公英提取物中绿原酸含量的测定。

【Abstract】 Objective:To establish an HPLC method for the determination of chlorogenic acid in dandelion extract.Methods:Agilent Zorbax SB C18 column(4.6 mm×250 mm,5 μm) was adopted;The mobile phase consisted of acetonitrile-0.4% phosphoric acid solution(10∶90)at a flow rate of 1.0 mL·min-1;The detection wavelength was set at 324 nm,and column temperature was 30 ℃.Results:The linear ranges of chlorogenic acid was 0.040-0.60 μg(r=0.9992);The average recoveriey(n=5) was 96.48%.Conclusion:HPLC determination of chlorogenic acid in dandelion extract is rapid,simple and accurate.It can be applied to determine of chlorogenic acid in dandelion extract.
