分光光度法测定阿奇霉素晶体的含量
Spectrophotometry determination of the contents of azithromycin crystal
【摘要】 目的:建立显色反应分光光度法测定阿奇霉素晶体的含量。方法:利用硫酸与阿奇霉素酮基质子化可发生显色反应,该反应溶液在483nm处具有最大吸收值。可用分光光度法对阿奇霉素进行定量分析,并采用正交实验设计优化了测定方法。结果:阿奇霉素浓度在18~72μg·mL-1范围内,显色溶液吸收度同阿奇霉素浓度具有良好的线性关系,线性回归方程为C(μg·mL-1)=-12.01+85.27A,r=0.9997,平均回收率为99.1%。结论:该方法简单、灵敏、准确,适合于阿奇霉素晶体含量的快速测定。
【Abstract】 Objective:To establish a spectrophotometric method for the determination of azithromycin crystal,based on color reaction.Method:Based on the protonation reaction of sulfuric acid and ketone group of azithromycin,the maximum Absorption value was obtained at 483 nm.The quantitative analysis of azithromycin was carried by spectrophotometric method,which was optimized by orthogonal design.Results:The linear equation was C (μg·mL-1)=-12.01+85.27A,relation coefficient was 0.9997 in the range of 18-72 μg·mL-1.The average recovery was 99.1%.Conclusion:The method is simple,sensitive,accurate,and suitable for the rapid determination of azithromycin crystal.
