辛伐他汀对牙周韧带细胞增殖和分化能力影响的实验研究

【摘要】 目的:探讨辛伐他汀对牙周韧带(PDL)细胞增殖和分化能力的影响,为辛伐他汀用于牙周组织再生提供理论依据。方法:组织块法培养人牙周韧带细胞,采用MTT法和二辛可宁酸法分别检测不同浓度辛伐他汀对牙周韧带细胞增殖能力和分化功能的影响。结果:高浓度辛伐他汀对PDL细胞增殖活性有轻微抑制作用,低浓度的辛伐他汀对PDL细胞增殖能力的影响不明显,无统计学意义(P>0.05)。但辛伐他汀可以明显促进PDL细胞中碱性磷酸酶的活性,尤其以10-7mol/L浓度的辛伐他汀对ALP活性的促进作用最明显,结果具有统计学意义(P>0.05)。以10-7mol/L浓度为中心,随着辛伐他汀浓度增加或减少,其促进碱性磷酸酶(ALP)活性的作用逐渐减弱。结论:辛伐他汀可以促进PDL细胞的骨向分化,可能对牙周组织缺损的再生具有重要意义。

【Abstract】 AIM:To study the effects of simvastatin on the functions of proliferation and differentiation of human periodontal ligament cells,and provide theoretical basis for periodontal tissue regeneration.METHODS: The periodontal ligament cells were cultured by explant culture technique.The proliferation and differentiation effect of simvastatin on PDL cells were investigated using MTT method and bicinchoninic acid method.RESULTS:High concentration of simvastatin has a slight inhibition on the proliferative activity of PDL cells,Low concentrations of simvastatin on the proliferative ability of PDL cells was not obvious.There was no statistical significance.However,simvastatin can promote the activity of alkaline phosphatase of PDL cells particular in 10-7mol/L concentration.There had obvious difference between groups.Taking the 10-7mol/L concentration as the center,with the increase or decrease in concentration of simvastatin,its function of promoting ALP activity was weakened gradually.CONCLUSION: Simvastatin can impel parts of periodontal ligament cells differentiating into osteogenesis.
