RP-HPLC法测定不同方法炮制关木通中马兜铃酸A的含量
RP-HPLC Determination of Content of Aristolochic Acid A in Different processing with Manchurian Dutchmanspipe Stem
【摘要】 目的:建立反相HPLC法测定不同方法炮制关木通中马兜铃酸A的含量。方法:采用反相高效液相色谱法(RP-HPLC),具体色谱条件:柱子:Cl8色谱柱(200mm×4.6mm,5μm);流动相:甲醇-2%冰醋酸梯度洗脱;检测波长:319nm;流速:l.0mL.min-1。结果:马兜铃酸A浓度在0.0046mg.mL-1~0.0002875mg.mL-1范围内(r=0.9999)呈良好的线性关系。结论:关木通经炮制后马兜铃酸含量均有不同程度降低(生品峰面积值:5374211,炮制品峰面积值:1632548~6027894),以碱制(峰面积值为1635145)醋炙(峰面积值为5059233),效果最为显著。同时比较浸出物含量,醋炙法保留浸出物含量效果较好,与生品相似,为最好的脱毒存效炮制方法。
【Abstract】 Objective:To set up a reversed-phase HPLC method for the determination of aristolochic acid I in Caulis Aristolochiae Manshuriensis.Methods:A reversed-phase HPLC method is applied.Specific chromatographic condition:Cl8 chromatographic column(200 mm×4.6 mm,5 μm) Mobile phase:methanol-2%aceti applly the method of gradient elution.Detecting wavelengh:319nm;Flow rate:l.0 mL·min-1.Results:There was a good linear relationship over a range of 0.0046 mg·mL-1~0.0002875 mg·mL-1 foraristolochic acidI(r=0.9999).Conclusion:The contents of aristolochic acidI in Caulis Aristolochiae Manshuriensis were lost when the original herbs processed by different ways(peakarea:5374211,1632548-6027894),especially for the methods of processing herbswith alkali(peak area:1635145) and ginger(peak area:5059233).
