雌二醇对去卵巢大鼠心肌和血管内皮细胞功能的影响

【摘要】 目的本实验用去卵巢大鼠建立更年期动物模型,给予不同剂量外源性雌二醇(E2),探讨E2对去卵巢大鼠心肌细胞和血管内皮细胞保护作用及机制。方法用RIA测定血清E2浓度和内皮素(ET)浓度;用酶法检测血清中NO浓度。用免疫组织化法检测心血管细胞雌激素受体的ER mRNA表达的观察。结果血清中NO与E2补充量呈正相关,ET与E2补充量呈负相关。ER mRNA在OVX组大鼠心肌细胞的胞浆中呈弱阳性表达,补充大剂量组大鼠心肌细胞的胞浆呈阳性表达。结论去卵巢后补充E2可使心血管中ER表达增加,提示绝经后心血管中ER含量减少可能是导致绝经后女性心血管病发病率迅速上升的重要机制之一。去卵巢后补充E2可使血清中NO含量增加,ET含量减少,说明血中E2可以影响或调节心血管内皮细胞分泌NO、和ET的含量。

【Abstract】 Objective The ovariectomized rats were used for climacteric period animal model and different dosages of extraneous estradiol(E2) were supplied.The protective function and the mechanism of E2 to the cardiac muscle cell and the blood vessel endothelial cell of the ovariectomized rats were discussed.Methods The examination of E2 and ET density in blood;the examination of nitrogen monoxide(NO)density in blood.Immunity to chemistry in dicatrix examination:the observation of expression of the messenger of the cardiovascular cell estrin receptor ribonucleic acid(RNA)(ER mRNA).Result The concentration of serum NO was positive correlation to amount of supplying estrogen, but the concentration of NO and ET was negative correlation.The expression of mRNA of estrogen receptor(ER mRNA) showed weakly positive the cardiac muscle plasma of ovariectomized rats, but which showed positively in the supplied large dose estrogen rats.Conclusions supplying estrogen can increase express of ER that maybe indicate the increase of incidence of disease of cardiovascular system in menopause women related to the lessening of ER.The concentration of NO is increased and that of ET is reduced in supplied estrogen ovariectomized rats;this shows exogenous estrogen in blood can affect or regulate the concentration of NO and ET, which is secreted by cardiovascular endothelial cell, but the dose of the supplied estrogen should be paid more attention.
