siRNA抑制BCR-ABL基因诱导K562细胞凋亡的初探
Exploration into the restraint of siRNA on the apoptosis of K562 cells induce by BCR-ABL genes
【摘要】 目的:通过mRNA结构预测软件预测出BCR-ABLmRNA二级结构,未形成二级结构的区域作为靶序列。方法:针对BCR-ABLmRNA二级结构中的单链区域设计四对siRNA,通过Lipotap脂质体作为转染载体去转染K562细胞。结果:所设计的四对siRNA其中有一对siRNA转染K562细胞后,K562细胞出现凋亡现象。结论:通过该方法有针对性的设计siRNA,避免了把mRNA的二级结构区域作为靶序列,提高了所设计siRNA的效果。
【Abstract】 Purpose: to predict BCR-ABLmRNA second-level structure through mRNA structure prediction software and with the area which has not formed second-level structure as target sequence.Method: according to the single chain area in BCR-ABLmRNA second-level structure,designing four pairs of siRNA and transferring K562 cells with Liptotap liposome as transfer vector.Result: among four pairs of siRNA,only one pair caused the apoptosis of K562 cells after transferring them.Conclusion: by designing siRNA,avoiding taking the second-level structure of mRNA as the target sequence and improving the effect of sinRNA.
