用条件性RNAi敲低技术降低FGFR2在小鼠软骨细胞中表达水平的研究
Research on the Conditional Knockdown of FGFR2 in Cartilag with RNAi Technology
【摘要】 目的:把条件性FGFR2敲低小鼠FGFR2-RNAi和在软骨细胞特异表达Cre重组蛋白的转基因小鼠Coll2a-Cre交配,得到双基因小鼠FGFR2RNAi;Coll2a-Cre,观察能否用条件性RNAi敲低的方法在小鼠软骨组织中降低FGFR2的表达,为进一步研究小鼠软骨中FGFR2表达降低对软骨发育的影响提供初步基础。方法:运用RT-PCR技术来比较小鼠软骨中FGFR2mRNA丰度的变化。结果:在双基因小鼠FGFR2RNAi;Coll2a-Cre软骨中FGFR2mRNA丰度同对照型相比表现为下降。结论:双基因FGFR2RNAi;Coll2a-Cre小鼠比同组对照小鼠软骨中FGFR2mRNA丰度要低,提示可用条件性RNAi敲低技术降低FGFR2在软骨中的表达,从而进一步观察软骨的发育情况。
【Abstract】 Objective to further investigate whether the decrease of FGFR2 in cartilage has an effect on the development of cartilage,the FGFR2 expression of the offspring FGFR2RNAi;Coll2a-Cre mice from FGFR2RNAi mice mating with Coll2a-Cre mice specific expressing Cre recombinant protein in cartilage are observed with conditional RNAi knockdown technology.Method Comparing the content of FGFR2mRNA in cartilage of FGFR2RNAi;Coll2a-Cre mice and controls by utilizing RT-PCR.Result the content of FGFR2mRNA in cartilage of FGFR2RNAi;Coll2a-Cre mice lower than that of controls.Conclusion The content of FGFR2mRNA in cartilage of FGFR2RNAi;Coll2a-Cre mice is lower than that of controls with RT-PCR.
