室旁核内iGluRs对旋转刺激诱发的大鼠心血管反应的影响

【摘要】 [目的]探讨下丘脑室旁核(PVN)内的促离子型谷氨酸受体(iGluRs)对旋转刺激诱发的心血管反应的影响.[方法]以旋转刺激为应激刺激,在电脑控制的代谢笼内记录大鼠清醒状态、自由活动时的心血管活动变化情况的同时,分别向PVN局部灌流2种离子型谷氨酸受体阻断剂,观察其对旋转刺激诱发的心血管反应的影响.[结果]向PVN区灌流N-甲基-D-天冬氨酸(NMDA)受体阻断剂MK-801,再给予旋转刺激,大鼠血压从(99.9±0.5)mmHg上升至(104.5±1.0)mmHg,心率从(357.3±9.1)次/min加快至(387.2±8.5)次/min,MK-801旋转组大鼠血压及心率的增加反应与对照组相比较明显受到抑制;向PVN区灌流非NMDA受体阻断剂CNQX,再给予旋转刺激,大鼠血压从(104.9±2.3)mmHg上升至(113.2±1.3)mmHg,心率从(375.2±5.7)次/min加快至(421.1±5.4)次/min,CNQX旋转组大鼠心血管反应未受到明显的影响.[结论]PVN内NMDA受体参与旋转刺激诱发的心血管反应的调节过程.

【Abstract】 OBJECTIVETo investigate the possible involvement of ionotropic glutamate receptors(iGluRs) in the PVN on cardiovascular responses induced by swing rotation.METHODSAntagonists of two kinds of iGluRs were perfused in the PVN region,then the cardiovascular responses induced by swing rotation were measured.RESULTSMK-801,an antagonist of NMDA receptor,was directly perfused in the PVN and swing rotation was added.The swing rotation increased the BP and HR from(99.9±0.5)mmHg and(357.3±9.1)beats/min to(104.5±1.0)mmHg and(387.2±8.5)beats/min;MK-801 attenuated the swing rotation-induced increases of BP and HR.CNQX,an antagonist of non-NMDA receptor,was directly perfused in the PVN and swing rotation was added.The swing rotation increased the BP and HR from in(104.9±2.3)mmHg and(375.2±5.7)beats/min to(113.2±1.3)mmHg and(421.1±5.4)beats/min;CNQX did not affect the cardiovascular responses induced by swing rotation.CONCLUSIONNMDA receptor in the PVN may be involved in the central regulation of cardiovascular responses induced by swing rotation.
