靶向超声造影微泡的制备及体外靶向性研究

【摘要】 目的制备一种靶向超声造影微泡,并进行体外靶向黏附效能评价。方法以磷脂为球壁材料,全氟丙烷为被包裹气体,采用高速剪切工艺制备氟碳脂质微泡,再应用生物素-亲和素桥连技术构建携带抗P-选择素单抗靶向氟碳脂质微泡,并应用P-选择素Fc段包被的平行板流动腔评价其体外靶向黏附效能。结果制备的靶向微泡呈圆整球形,粒度分布均匀,体外靶向黏附作用明显。结论本试验工艺可成功制备靶向氟碳脂质超声造影微泡,平行板流动腔法有助于评价靶向微泡的靶向黏附效能。

【Abstract】 Objective To prepare a targeted ultrasound contrast agent,perfluorocarbon-filled lipid microbubbles,and investigate its in vitro targeting binding capability.Methods The agitation of high-speed method was employed to prepare the perfluorocarbon-filled lipid microbubbles,and biotin-streptavidin-mediated technique was used for the attachment of anti-mouse P-selection monoclone antibody to the performed microbubble shell. An in vitro targeting assay was performed using parallel plate flow chamber.Results The targeted microbubbles were spherical and well-distributed.In vitro targeting test showed that these microbubbles can selectively attached to receptor-coated surfaces.Conclusion This method can be applied to the preparation of targeted ultrasound contrast agents. Parallel plate flow chamber experiment can be used to assess the binding capability of targeted microbubbles.
