交感神经递质去甲肾上腺素对肝细胞内核转录因子活化的调控
Effects of norepinephrine on the activity of nuclear factors in hepatocytes
【摘要】 目的了解交感神经递质去甲肾上腺素对肝细胞内核转录因子NFκB p50/p65及其配体IκBα的表达变化的调控。方法应用终浓度为10μmol/L的去甲肾上腺素作用于Wistar大鼠原代培养肝细胞,作用时相点为5、15、30和60 min。收集细胞提取细胞核蛋白,采用EMSA法检测NFκB p50/p65的变化。同样以Western blotting法进行IκBα含量的检测。结果由EMSA电泳图像中可见NE作用5分钟时即可见到有NFκB p50/p65明显活化,并随时间增强而活化逐渐明显。计算灰度面积值在5 min后各测得值均较对照组明显增高(P<0.01)。NE作用肝细胞5 min后,IκBα表达量明显减少,刺激30~60min时只能检测到其微量表达(P<0.01)。结论NE可导致内皮细胞NFκB p50/p65发生核移位,并使IκBα降解,进而活化NFκB,诱导肝细胞分泌细胞因子。
【Abstract】 Objective To study the effects of norepinephrine(NE) on the activity of nuclear factors in hepatocytes.Methods Primary cultured hepatocytes of Wistar rats were administered with NE(10 μmol/L) for 5,15,30 and 60 minutes respectively.The activity of NFκB was measured by EMSA method in each group.Meanwhile,IκB was detected by Western blotting.Results NE increased significantly the levels of NF-κB activity 5 min after administration,and forward gradually at 15,30 and 60 min(P<0.01).On the contrary,IκB degraded markedly at 5 min after adminstrtion(P<0.01), and only minimal activity of IκB could be detected at 30 and 60 min.Conclusion NE might induce the nuclear translocation of hepatocytes NFκB p50/p65 and IκBα degradation and activate NFκB,which ultimately leads to the secretion of cytokines from the hepatocytes.
