川芎幼嫩叶片植株再生的研究

【摘要】 为了建立一套川芎Lisgusticum chuanxiong Hort快速繁殖技术。以川芎幼嫩叶片为外植体,以MS和1/2MS为基本培养基,通过研究6-BA、2,4-D、NAA、IAA、和IBA对愈伤组织诱导、不定芽分化和不定根产生的影响,找出相应的最佳激素组合。结果表明:川芎最佳愈伤组织培养基为MS+6-BA0.5mg·L-1+2,4-D2mg·L-1;最佳不定芽分化培养基为MS+6-BA0.5mg·L-1+NAA0.5mg·L-1;最佳生根培养基是1/2MS+NAA0.5mg·L-1+IBA0.5mg·L-1。组培苗移栽后,生长健壮,成活率达100%。

【Abstract】 To establish a protocol of rapid clonal propagation of Lisgusticum chuanxiong Hort,using immature leaves as explants.Tthe optimal hormone combinations for callus induction,adventitious bud initiation and rooting were found out by using MS and 1/2 MS medium and the effects of 6-BA,2,4-D,NAA,IAA and IBA on the processes were studied.Result and Conclusion:MS+ 6-BA 0.5 mg·L-1+2,4-D 2 mg·L-1 was the best medium for calli initiated ;MS+ 6-BA 0.5 mg·L-1+ NAA 0.5 mg·L-1 was the best medium for adventitious bud initiated ;1/2MS+NAA0.5 mg·L-1+IBA 0.5 mg·L-1 was the best medium for rooting induction.The regenerated plantlets grew vigorously after they were transplanted and the survival rate was 100% 。
