利用白腐真菌对柑橘皮渣进行预处理及厌氧发酵产甲烷研究

【摘要】 从柑橘果园的堆积土中筛选出一株丝状白腐真菌,通过正交试验确定其最佳的生长环境并粗略测定其酶活力,利用该菌株对柑橘皮渣进行分解,并对分解后的柑橘皮渣进行厌氧发酵产甲烷试验.研究表明:在该菌株单因素下,25天内,该菌株对柑橘皮渣的分解率平均达到30%.正交实验结果显示,对柑橘皮渣分解最好的处理组合条件是麸皮2g、柑桔皮渣含水量60%、初期pH3、硫酸铵0.2g.产生还原糖量平均达到517μg/g,木糖120.8μg/g,纤维素+纤维素分解量0.96g,均失重达到67%,能够有效提高柑橘皮渣的pH从pH1提高到pH4.酶活力测定表明,菌丝覆盖柑桔皮渣的第8天到第26天之间,AVI的活力均要远高于CMC和FPA.AVI在培养的第13天酶活力达到最大,第17天后酶活性直激下降.其中CMC在整个培养期间几乎没有变化,一直保持平稳.对柑橘皮渣预处理后进行厌氧产甲烷发酵,试验表明,处理后的柑橘皮渣产沼气总产量较不处理前提高20%,甲烷含量提高26%,COD值都能有效地降低达到27%,发酵前后还原糖量降低20%.

【Abstract】 A filiform white-rot fungus was screened from the soil pile of a citrus orchard.An orthogonal test was conducted to determine the optimum environment for its growth and to roughly estimate its enzyme activity.Then this fungus strain was used to decompose citrus fruit dregs,and the decomposed citrus dregs were made to ferment anaerobically to produce methane.The results indicated that under the single factor of this strain,30%,on the average,of the citrus dregs were decomposed in 25 days.The optimum combination of conditions for decomposition was bran 2 g,water content of the citrus peel dregs 60%,initial pH 3 and ammonium sulfate 0.2 g.Under these conditions,the reducing sugar produced averaged 517 μg/g;xylose 120.8 μg/g;cellulose + cellulose decomposition was 0.96 g;weight loss averaged 67%;and the pH of the citrus fruit dregs was enhanced from 1 to 4,thus preparing an advantageous condition for the next step of anaerobic fermentation.The enzyme vitality test indicated that the AVI vigor was significantly higher than that of CMC and FPA from the 8th day to the 26th day of covering citrus peel dregs with the hyphae.The activity of AVI reached the maximum on the 13th day of culture and sharply dropped after the 17th day.In contrast,CMC showed very little change throughout the culture period.Pretreatment before anaerobic fermentation increased biogas production by 20% and methane content by 26%,and decreased COD value by 27% and reducing sugar by 20%.
