降低西瓜试管苗玻璃化因素的优化
Optimization of Factors for Decreasing Vitrification of Watermelon Plantlets in vitro
【摘要】 以西瓜为材料,采用L27(313)多因素正交设计,研究不同的基因型材料、培养温度、光照强度、培养基中BA、琼脂、蔗糖、IAA和Phl.浓度对试管苗玻璃化的影响.结果表明:培养温度的影响达极显著水平(p<0.01),玻璃化频率15℃>35℃>25℃,15℃与25℃、35℃间差异显著(p<0.05);材料的基因型、光照强度、培养基中BA、琼脂和蔗糖浓度的影响达显著水平,玻璃化频率2n>3n>4n,4n与2n、3n间差异显著;1 000 Lx>2 000 Lx>3 000 Lx,1 000 Lx与2 000、3 000 Lx间差异显著;BA 4 mg/L>2 mg/L>1 mg/L,1 mg/L与2 mg/L、4 mg/L间差异显著;琼脂5 g/L>8 g/L>11 g/L,5 g/L与8 g/L、11 g/L间差异显著;蔗糖10 g/L>30 g/L>50 g/L,10 g/L与30 g/L、50 g/L间差异显著.在继代培养中,降低试管苗玻璃化的综合措施是:培养温度25~35℃,光照强度2 000~3 000 Lx,培养基中BA≤1 mg/L,IAA≥1 mg/L,Phl.≥2 g/L,琼脂8~11 g/L,蔗糖30~50 g/L.
【Abstract】 Watermelon varieties Zhengkong No.1(2n),Zhengkong seedless No.1(3n) and Tetraploid No.1(4n) were used as materials in an experiment of orthogonal design L27(313).The factors studied were the genotype of the material(A),culture temperature(B),6-BA(C),IAA(D),agar(E),light intensity(F),sucrose(G) and phloridzin(Phl.,H).Buds coming from different genotype materials were inoculated as explants on MS medium supplied with different concentrations of agar,sucrose,6-BA,IAA and Phl.(pH=5.8) after they were sterilized,and cultured under different temperature and light conditions for 30 days.The results showed that factor B influenced vitrification of explants highly significantly(p<0.01),factors A,C,E,F and G influenced vitrification of explants significantly(p<0.05),and there were significant differences among different levels of the above factors at 0.05 level.In order to prevent or decrease vitrification of watermelon plantlets in vitro,culture temperature should be controlled in the range from 25 to 35 ℃,light intensity from 2 000 to 3 000 lx,the concentration of 6-BA in MS medium ought to be below 1 mg·L-1,and the concentration of agar and sucrose in medium should be from 8 to 11 g·L-1 and from 30 to 50 g·L-1,respectively.
