陇东绒山羊微卫星DNA遗传多样性的研究

【摘要】 利用9个微卫星座位,8%非变性聚丙烯酰胺凝胶电泳PCR扩增产物,对120只陇东绒山羊基因组DNA的遗传多样性进行了检测,对其群体遗传学特性进行分析。结果表明:陇东绒山羊在9个微卫星座位中共检测到90个等位基因,其中微卫星座位LSCV24等位基因数最多,为14个;BMS1248多态信息含量最高,PIC为0.88;DB6座位多态信息含量最低,PIC为0.79。在所标记群体中平均有效等位基因数为7.69±1.65个,平均多态信息含量为0.84±0.03;遗传杂合度均值为0.86±0.03,均高于国内外部分研究结果。说明陇东绒山羊品种遗传多样性丰富,群体遗传变异程度高,育种潜力大。

【Abstract】 The genetic polymorphisms of 9 microsatellite loci in 120 cashmere goats of East Gansu Province,a special chinese genetic resource,were studied to reveal genetic diversity of the population by 8% non-denatured polyacrylamide gel electrophoresis detecting the product of PCR amplifying and allele frequencies,Heterozygosities and PIC values calculated.The results showed that: the total number of alleles were 90,and allele numbers of LSCV24 that were 14 were the most,Polymorphic information contents of BMS1248 were the highest,PIC was 0.88and that of DB6 were the lowest,PIC was 0.79 at nine microsatellite loci in Cashmere goats of East Gansu Province.The average effective number of allele,the average PIC values and the average genetic heterozygosity were 7.69±1.65、0.84±0.03 and 0.86±0.03 respectively,which were higher than that of others.It showed that genetic Polymorphism of cashmere goats of East Gansu Province was very rich,Genetic Variation degree of the breed was high,and breeding potential was great also.
