ezrin和AKT-1在眼表鳞状细胞肿瘤中的表达
Expression of ezrin and AKT-1 in squamous carcinoma at ocular surface
【摘要】 目的观察ezrin和AKT-1在眼表鳞状细胞肿瘤中的表达情况,了解这两种分子与眼表鳞状细胞肿瘤发展和转化的关系、与肿瘤细胞增殖活性的关系,以及两种分子作用的相互关系。方法采用苏木素伊红染色观察65例眼表鳞状细胞肿瘤的组织形态学特征,根据肿瘤细胞异型性和肿瘤有无浸润将65例病例分为3组(鳞状细胞乳头状瘤组22例,鳞状细胞乳头状瘤伴有非典型增生组18例,鳞状细胞癌组25例)。用免疫组织化学染色检查各组中ezrin、AKT-1和Ki67的表达情况并进行统计学分析。结果ezrin在鳞状细胞癌组的表达(23/25)明显高于鳞状细胞乳头状瘤组(7/22)和鳞状细胞乳头状瘤组伴有非典型增生组(5/18)(均为P<0.05),但在两种乳头状瘤之间表达无明显差异。AKT-1表达在3组病例中均有明显差异(鳞状细胞癌组22/25,鳞状细胞乳头状瘤组5/22,鳞状细胞乳头状瘤伴非典型增生组13/18);ezrin和AKT-1在两种鳞状细胞乳头状瘤中均无明显的共表达(鳞状细胞乳头状瘤4/22,鳞状细胞乳头状瘤伴非典型增生5/18),而在鳞状细胞癌中二者共表达则明显增多(20/25,P<0.05);在Ki67表达≤15%病例中(37例)和>15%病例(28例)之间:ezrin表达有明显差异(10/37,25/28;P<0.05),而AKT-1表达差异则无统计学意义(22/37,18/28;P>0.05)。结论ezrin和AKT-1的表达与眼表鳞状细胞肿瘤的恶性转化可能相关,ezrin的作用可能与AKT-1的表达有关。ezrin对眼表鳞状细胞肿瘤的肿瘤细胞增殖活性有明显影响。
【Abstract】 Objective To investigate the correlation of ezrin and AKT-1 to the development and transformation of squamous carcinoma at ocular surface,and to comprehend their correlation to proliferative ability of tumor cells and interrelation between ezrin and AKT-1 by observing the expression of them in squamous carcinoma at ocular surface.Methods Histomorphological characteristics of 65 cases with squamous carcinoma at ocular surface were observed by hematoxylin and eosin staining.A total of 65 cases were divided into 3 groups with reference to heteromorphism and with or without infiltration of tumor cells.There were 22 cases in squamous papilloma group,18 cases in squamous papilloma combined with atypical hyperplasia and 25 cases in squamous carcinoma group.Immunohistochemical staining was used to measure the expression of ezrin,AKT-1 and Ki67 in different groups and the results were statistically analyzed.Results There was a higher expression of ezrin in squamous carcinoma(23/25)than that in common squmaous papilloma(7/22)and that in squamaous papilloma combined with atypical hyperplasia(5/18)(both P<0.05),but the difference was significant between two kinds of squmaous papillomas.The expression of AKT-1 has an obvious difference among the cases of three groups(squamous carcinoma with 22/25,common squmaous papilloma with 5/22,squmaous papilloma with atypical hyperplasia with 13/18);There was a statistical difference in the expression of ezrin between the cases with the expression of Ki67 less than or equal to 15%(37 cases)and those with expression of Ki67 more than 15%(28 cases)(10/37,25/28;P<0.05),but the expression of AKT-1 was not significant(22/37,18/28;P>0.05).The coexpression of ezrin and AKT-1 was not obvious in two kinds of squmaous papilloma(4/22 in common squmaous papilloma;5/18 in squmaous papilloma with atypical hyperplasia),but increased obviously in squmaous carcinoma(20/25 in squamous papilloma,P<0.05).Conclusions The expression of ezrin and AKT-1 may affect malignant transformation of squamous carcinoma at ocular surface.The effect of erizen maybe associate with the expression of AKT-1,and ezrin can influence the proliferation of tumor cells in squamous carcinoma at ocular surface.
