氟砷单独及联合对正常人淋巴细胞增殖及IL-2分泌情况的影响观察

【摘要】 目的:观察氟、砷单独及联合作用对正常人淋巴细胞的增殖及IL-2分泌情况,为探讨氟、砷单独及联合作用对机体免疫功能的影响规律及其作用机制提供科学依据。方法:采用体外细胞培养,应用四甲基偶氮唑盐比色法(MTT法)、酶联免疫吸附试验(ELISA)法,观察0.1、1.0μmol/L亚砷酸钠(NaAsO2)及10、50μmol/L氟化钠(NaF)对正常人淋巴细胞的生长抑制及细胞因子分泌的影响。结果:人淋巴细胞在经浓度为0.1、1.0μmol/L的NaAsO2及浓度为10、50μmol/L的NaF染毒后,其MTT吸光度值与对照组相比,均呈下降趋势,且有统计学意义(P均<0.05);浓度为1.0μmol/L的NaAsO2及浓度为50μmol/L的NaF与淋巴细胞作用,可抑制白细胞介素-2(IL-2)的分泌,但未发现氟砷联合作用对人淋巴细胞的作用呈现交互作用(P>0.05)。结论:氟、砷对人淋巴细胞存在明显的生长抑制作用,并与NaAsO2及NaF在体外对人淋巴细胞IL-2分泌的抑制作用结果相吻合。提示氟、砷对免疫细胞的生长抑制作用可能是砷、氟对机体免疫系统损伤的重要因素之一。

【Abstract】 Objective: To investigate the biological effect and function mechanism of sodium arsenite(NaAsO2) and sodium fluoride(NaF) in human lymphocyte in vitro and provide the basis for further study.Methods: The actions of NaAsO2 and NaF on the cell growth and interleukin-2(IL-2) secreting in human lymphocyte in vitro were observed by MTT assays and enzyme linked immunosorbent assay.Results: It showed from our study that NaAsO2 and NaF had a remarkable inhibitory effect on the proliferation of human lymphocytes with the concentration of 1 μmol/L and 50 μmol/L respectively(P<0.05).The concentration of 1 μmol/L NaAsO2 and 50 μmol/L NaF showed significant inhibition on IL-2 secreting(P<0.05).The interaction of NaAsO2 and NaF was not found in this test(P>0.05).Conclusion: Both arsenic and fluoride have a poisoning effect on human lymphocytes with lower dose,but the interaction in the combination group was not found,it was simply additive.
