反相高效液相色谱法测定人尿拉米夫定浓度
Determination of lamivudine in urine with RP-HPLC
【摘要】 目的建立测定人体口服阿米洛利尿液中含量的方法。方法采用RP-HPLC法,色谱条件为Gemini C18柱(250 mm×4.6 mm,5μm);流动相0.02 mol/L磷酸盐缓冲液-甲醇(90:10);流速1.0 ml/min;检测波长274nm;柱温30℃。结果拉米夫定在0.0067 mg/ml~0.0733mg/ml浓度范围内线性关系良好(r=0.9997),加样回收率分别为96.00%、94.00%、97.67%,RSD为1.91%(n=5)。结论该方法能灵敏、准确的测定尿液中拉米夫定的含量。
【Abstract】 Objective To establish a method to measure lamivudine in urine.Method A method of RP-HPLC was established,whose parameters were included Gemini C18 column(250 mm×4.6 mm,5 μm),mobile phase(0.02 mol L-1 phosphate buffer,-CH3OH(90:10)),flow rate(1.0 ml min-1) detective wavelength(274 nm) and temperature of column(30℃).Results The linear range was 0.0067 mg ml-1~0.0733mg ml-1(r= 0.9997),with the average recovery rate of 96.00%,94.00,97.67%,and RSD of 1.91%(n=5).Conclusion The concentration of lamivudine in urine can be determined sensitively and accurately by RP-HPLC.
