【摘要】 目的探索成年糖尿病大鼠心室肌细胞成功分离的方法。方法取体质量150~200 g的Sprague-Daw ley大鼠20只,单次腹腔注射链脲佐菌素(STZ)建立DM大鼠模型,3周后取大鼠心脏行主动脉逆行灌流分离心室肌细胞;并在倒置显微镜下观察分离的单个心室肌细胞状态。结果建模成功率达90%,糖尿病大鼠的血糖水平稳定维持在20 mmol/L以上;分离的单个心室肌细胞存活率达70%~90%,且纹理清晰状态好。结论该糖尿病心室肌细胞的分离方法简单有效。更多还原

【Abstract】 AIM To explore an effective method for successful isolation of ventricular myocytes of adult diabetic rats.METHODS The diabetic rat model was established by a single injection of streptozocin(STZ) peritoneally in Sprague-Dawley rats weighing 150-200 g.Ventricular myocytes were isolated by perfusion of enzymatic solution.The isolated ventricular myocytes were then observed under inverted microscope.RESULTS The successful establishment rate of diabetic rats was 90%,the blood glucose level stayed high above 20 mmol/L.The isolated ventricular myocytes were alive up to 70%-90% with good and clear stripes.CONCLUSION This simple method is an effective way for successful isolation of diabetic rat ventricular myocytes.更多还原
