去乙酰真菌环氧乙酯琼脂表面培养基质的优化

【摘要】 去乙酰真菌环氧乙酯是一种骨架为含氧桥的环二烯类化合物,具有较高的抗肿瘤活性.以其产生菌拟茎点霉414菌株(Phomopsissp.414)出发,利用响应面法对琼脂表面培养的发酵基质进行优化.采用单因素法和Plackett-Burman设计,从17种因素中筛选出提高产量的主要因素,继而利用响应面法确定各因素的最优水平.当葡萄糖为25.56 g/L、甘露醇为12 g/L、马铃薯为231.82 g/L时,拟茎点霉414菌株发酵产去乙酰真菌环氧乙酯可达108.9 mg/L,为优化前的3.2倍.

【Abstract】 Because of the advantages of new mechanisms,high anti-tumor activity,low side effects and new structure,deacetylmycoepoxydiene has been the candidate of new drugs.Deacetylmycoepoxydiene is a second metabolic product of Phomopsis sp.414,containes an oxygen-bridged cyclooctadiene.Deacetylmycoepoxydiene has high anti-tumor activity.The IC50 of Raji cell line is 3.0 μg/mL.But the further research is hindered for its low yield.This paper successfully solves the problem in solid fermentation.By response surface method,the deacetylmycoepoxydiene yield of Phomopsis sp.414 increases considerably.Based on the result of one factor experimental design,nine key factors with strong impact on deacetylmycoepoxydiene,namely,glycerine,glucose,manicol,starch,potatoes,cyclohexanhexol,micotinamide,vitamin B and folie acid were selected.For reducing the error,the last four factors are united into multi-vitamin.By Plackett-Burman design,the six factors are been measured by t value.Mannitol is been identified as the most important factor.Then the basic factors of PDA and the most important factor mannitol were further investigated with Box-Behnken and response surface analysis.The results show that mannitol 12 g/L,glucose 25.56 g/L,potatoes 231.82 g/L,the production of deacetylmycoepoxydiene reaches to 108.9 mg/L,3.2 times as before.This experiment lays the foundation for the follow-up development of deacetylmycoepoxydiene.
