老龄骨质疏松骨形态、骨密度、生物力学实验研究

【摘要】 [目的]观察自然衰老骨质疏松模型中雌性大鼠骨形态、骨密度、骨生物力学改变,探索老年妇女骨质疏松症发生发展的机制。[方法]选择健康纯种雌性SD大鼠10只,喂养22月后复制自然衰老骨质疏松模型,另随机选择6月龄雌性大鼠10只为青年对照组,观察其骨形态、骨密度、骨生物力学指标。[结果]与青年对照组比,骨质疏松组骨小梁面积百分比显著降低(50%,P﹤0.05),股骨平均骨小梁数显著减少(65%,P﹤0.05),平均骨小梁宽度显著变窄(27%,P﹤0.05)。②骨质疏松组股骨与椎骨骨密度(64.3±8.1g/cm2,59.3±15.1g/cm2)较青年对照组(81.8±7.2g/cm2,92.2±6.1g/cm2)显著降低(P﹤0.05),椎骨骨密度下降得更明显(35.7%比21.3%)。③骨质疏松组股骨与椎骨的最大负荷力(134.41±23.52N,235.0±12.2N)较青年对照组(155.81±17.84N,327.1±24.8N)显著降低(P﹤0.05),且椎骨的最大负荷力降低更明显(28%比14%)。骨质疏松组椎骨、股骨的最大应变(1.28±0.12mm,1.6±0.15mm)与青年对照(1.38±0.15mm,1.81±0.21mm)比有明显下降的趋势,但尚未形成统计学差异。[结论]老年雌性大鼠骨质疏松症中骨形态发生、骨密度明显改变,骨形态学、变化更能反映早期的骨结构的变化,这种变化发展下去可能会影响骨的力学性能。

【Abstract】 [Objective] To observe bone martin,bone mineral density(BMD) and biomechanics changes in aged female rat,so as to explore mechanism in aged female osteoporosis [Methods] Female rats were divided into two groups:10 rats aged 22 months group were selected as the osteoporosis model group and the another 10 rats aged 6 months old were selected as the young control group.The bone martin,bone mineral density and biomechanics changes were observed.[Results] ① Comparing with six mouths old female rat,the bone trabeculae of volumes,the number,the width has significantly changed(50%,65%,27%,P﹤0.05).② Comparing with six mouths old female rat,BMD of femurs and Vertebra in the aged female osteoporosis rat significantly decreased(134.41±23.52N,235.0±12.2N,P﹤0.05).BMD of Vertebra had more changed than femurs.③ Comparing with six mouths old female rat(155.81±17.84N,327.1±24.8N),maxim stress of femurs and vertebra in the aged female osteoporosis rat were significantly decreased(134.41±23.52N,235.0±12.2N,P﹤0.05),and maximum load of vertebral obviously decreased(from 20% to 14%).The maximum strain of osteoporosis vertebrae(1.28 ±0.12mm and 1.6 ± 0.15mm) and femoral had obvious decreased when compared with youths(1.38±0.15mm and 1.81±0.21mm),but there was no significant difference between the two groups.[Conclusion] Bone,martin and BMD have significantly changed in female osteoporosis,indicating bone martin and BMD can reflect degeneration early.Changes in aged female osteoporosis,and this changes maybe influent bone biomechanics.
