实验性矽肺相关基因的表达谱研究
INVESTIGATION ON THE GENE EXPRESSION PRO FILE OF EXPERIMENTAL SILICOSIS
【摘要】 [目的]采用基因芯片技术探讨矽肺异常基因的表达。[方法]建立矽肺大鼠模型,提取矽肺组织和正常肺组织mRNA,分别用Cy5-dCTP或Cy3-dCTP标记,再与5705点基因芯片杂交,筛选矽肺组织与正常肺组织表达差异的基因。[结果]矽肺组织与正常肺组织表达差异基因323个,其中高表达的基因109个和低表达基因214个。[结论]基因芯片可快速筛选矽肺表达异常的相关基因,并有利于进一步探讨矽肺的发病机制。
【Abstract】 [Objective] To screen genes with abnormal expression in silicosis with cDNA chip. [Methods] The model of animal was established. The mRNA was isolated from silicosis and normal lung,and was labeled by Cy5-dCTP or Cy3-dCTP as probes. Then the mixed probes were hybridized to cDNA chip containing 5705 genes and genes with different expressions between silicosis and normal lung tissue were screened. [Results] 323 genes were differentially expressed between silicosis and normal lung tissue. Compared with normal lung tissue,109 genes were up-regulated and 214 genes were down-regulated in silicosis tissue. [Conclusion] The analysis of the gene expression profile of silicosis using gene chip can rapidly select associated abnormal genes,while further analysis of the obtained genes can help elucidate the pathogencity of silicosis.
