镀金和多壁碳纳米管修饰的金电极葡萄糖传感器

【摘要】 背景:目前报道的一些酶电极法葡萄糖传感器存在着诸如葡萄糖氧化酶易失活,稳定性和重复性较差等缺点,因此快速、稳定、准确的葡萄糖传感器已成为研究者关注的焦点。目的:构建一种新型的电流型电化学葡萄糖传感器。设计、时间及地点:观察实验,于2008-08/2009-02在解放军广州军区广州总医院医学实验科传感实验室内完成。材料:采用自组装技术将多壁碳纳米管固定到金电极上,再将纳米金电镀到金电极表面,最后通过静电作用吸附葡萄糖氧化酶,制成了电化学葡萄糖传感器。方法:以铁氰化钾作为探针,采用三电极系统循环伏安法和线性扫描法考察了传感器的组装特性,并观察不同修饰电极对葡萄糖响应峰电流的特性。主要观察指标:不同膜修饰电极的循环伏安特性;传感器对不同浓度葡萄糖的电流响应。结果:该传感器对葡萄糖浓度的检测具有优良的特性,线性检测范围是0.01~10mmol/L,检测限是0.4μmol/L,且具有响应速度快(<2s)、重复性及稳定性好等优点。将其用于临床血清检验,传感器的检测血糖值与己糖激酶法检测血糖值基本相符。结论:该传感器表现出优良的特性,响应速度快、稳定性和重现性好,可以用于对患者血糖浓度的临床检测。

【Abstract】 BACKGROUND: Recent reports indicate that enzyme-electrode glucose biosensor is limited by glucose-oxidase deactivation, poor stability and repeatability; therefore, glucose biosensor characterizing by rapid, stable, and exact arouses more and more attention. OBJECTIVE: To construct a novel electrochemical glucose biosensor. DESIGN, TIME AND SETTING: An observational experiment was performed at Bisensor Laboratory of Department of Medicine Laboratory, Guangzhou General Hospital of Guangzhou Military Command of Chinese PLA from August 2008 to February 2009. MATERIALS: The modified electrode was constructed, based on immobilization of glucose oxidase (GOD) on an Au electrode by electrostatic effect, which was modified with MWCNTs by utilizing self-assemble techniques and NanoAu by electrodeposition. METHODS: The ferricyanatum Kalium was used as probe. The properties of glucose biosensor were characterized by cyclic voltammetry (CV) and linear scan voltammetry (LSV) in the three-electrode system. Relation between glucose concentration and response peak current about different modified electrode were compared. MAIN OUTCOME MEASURES: The cyclic voltammetry of different modified electrodes; relation between glucose concentration and response peak current. RESULTS: Primary study indicated that the biosensor exhibited excellent performances for glucose detection with a low detection limit (0.4 μmol/L), a wide linear range (0.01-10 mmol/L), a fast response time (< 2 s), and good reproducibility and stability. In addition, the biosensor was applied in the determination of glucose in human blood samples and the results which were obtained from biosensor were agreement with Hexokinase (HK). CONCLUSION: The biosensors exhibit good reproducibility, stability and fast response to glucose, so it can be used in the clinic detection of glucose concentration in human serum.
