不同消毒方法对海藻酸钠-明胶共混体系理化性能的影响
Effect of different sterilization on sodium polymannuronate-gelatin complex
【摘要】 背景:不同的消毒方式对生物材料的理化性能均有一定的影响,影响的大小就成为选择消毒方法的重要参考依据。目的:观察不同的消毒方式对海藻酸钠-明胶共混体系理化性能的影响,并选择最佳的消毒方式。设计、时间及地点:观察对比实验,于2007-08在北京世纪坛医院中心实验室和中国石油大学油气储运工程系完成。材料:海藻酸钠干粉购自美国Sigma公司,明胶干粉购自河北绿岛公司。方法:分别以固体60Co照射消毒、固体高压蒸汽消毒和溶液高压蒸汽消毒后的40g/L海藻酸钠溶液与消毒明胶配制成海藻酸钠与明胶质量比为2∶3的海藻酸钠-明胶共混体系,终浓度为50g/L。磁力搅拌30min,静置30min后低速离心以使小气泡消失。以未消毒的海藻酸钠粉末与明胶配制的海藻酸钠-明胶共混体系为对照。主要观察指标:分别测定不同消毒方法消毒后的海藻酸钠-明胶共混体的pH值,吸光度值和黏度。结果:固体高压蒸汽消毒法、溶液高压蒸汽消毒法、60Co照射消毒法pH值分别为7.25,6.81,6.88;吸光度值分别为0.168,0.166,0.170;黏度分别为2.177,1.796,1.889Pa·s。结论:各种消毒方法对海藻酸钠的理化性能均有影响,其中,固体高压蒸汽消毒法影响相对移小。
【Abstract】 BACKGROUND:Different sterilization has different effect on physicochemical properties of biomaterials,therefore,effect degree provide a reference for selecting sterilization method.OBJECTIVE:To study effect of different sterilization on sodium polymannuronate-gelatinum complex and find out the optimal sterilization method.DESIGN,TIME AND SETTING:An observation and contrast experiment was completed at Center Laboratory of Beijing Shijintan Hospital and Department of Oil/Gas Storage and Transportation Engineering China University of Petroleum in August 2007.MATERIALS:Sodium polymannuronate powder was purchased from Sigma Inc.Gelatinum powder was supplied by Hengshui Lüdao Gelatin Co,.Ltd.METHODS:Sodium polymannuronate was sterilized using 60Co irradiation,high pressure vapor sterilization or solution vapor sterilization,respectively.Combined sterilized sodium polymannuronate with gelatinum at the mass ratio of 2:3,the terminal concentration was 50 g/L.Then magnetic stirring the sodium polymannuronate-gelatin complex for 30 minutes,and low-speed centrifugal was performed following standing for a second 30 minutes.Sodium polymannuronate with gelatinum which was without sterilization was served as control group.MAIN OUTCOME MEASURES:The pH value,A value and viscosity of sodium polymannuronate-gelatinum complex were measured after different sterilization methods were applied.RESULTS:The pH value of 60Co irradiation,high pressure vapor sterilization or solution vapor sterilization was 7.25,6.81,6.88,and A value was 0.168,0.166,0.170,with viscosity of 2.177,1.796,1.889 Pa·s,respectively.CONCLUSION:All sterilization methods have effect on sodium polymannuronate-gelatinum complex.The high pressue vapor sterilization has relatively little influence.
