雌、孕激素通过腺苷A2BR通路调节JEG-3绒癌细胞系上调MMP-2的表达
Upregulation of MMP-2 by progestogen and oestrogen via adenosine receptor A2BR signal pathway in JEG
【摘要】 目的:观察雌、孕激素对人绒癌细胞系JEG-3腺苷受体表达的影响,探讨它对JEG-3侵袭性的影响。方法:JEG-3细胞在含雌、孕激素环境中培养24h,设MRS1706(A2BR阻断剂)干预组及空白对照组;用明胶酶谱法检测MMP-2、MMP-9蛋白表达,RT-PCR技术检测JEG-3细胞腺苷受体mRNA水平。结果:RT-PCR结果显示,JEG-3细胞表达腺苷A1R、A2AR及A2BR3个亚型,未检测到A3R的表达;雌激素和(或)孕激素处理组A2BRmRNA含量显著高于对照组(P<0.05);明胶酶谱检测发现雌、孕激素联合处理显著上调JEG-3细胞MMP-2的分泌水平(P<0.05),但对MMP-9无显著影响;阻断A2BR信号途径显著下调JEG-3MMP-2表达,并能抑制雌、孕激素对MMP-2的上调效应(P<0.05)。结论:雌孕激素能改变JEG-3绒癌细胞腺苷受体的表达模式,呈A2BR优势表达,A2BR信号通路参与雌孕激素对JEG-3细胞侵袭性的调节。
【Abstract】 Objective:To study the effect of progestogen and/or oestrogen on expression of adenosine receptors and the ability of invasion in JEG-3.Methods:JEG-3 cell was cultured in progestogen and/or oestrogen condition for 24 hours with control or MRS1706(A2b-receptor specific antagonist),and MMP-2/MMP-9 in cultured supernatant was detected by zymogram.Adenosine receptors(A1R,A2AR,A2BR,A3R)was detected by employing semi-quantitative method of reverse transcription polymerase chain reaction(RT-PCR),and mRNA of A2BR was detected precisely by real time-PCR.Results:The result of RT-PCR showed expression of adenosine receptors(A1R,A2AR,A2BR) in JEG-3 without detecting A3R,and mRNA of A2BR was upregulated by progestogen and/or oestrogen compared with the control(P<0.05).The expression of MMP-2 was upregulated by progestogen and oestrogen,howerver,MMP-9 was not affected.Blockade of A2BR signal pathway in JEG-3 led to downregulation of MMP-2 and inhibited the upregulation of MMP-2 performed by progestogen and oestrogen with significance difference(P<0.05).Conclusion:Progestogen and/or oestrogen can change expression of adenosine receptors and expression of A2BR is predominate compared with others,thus A2BR signal pathway is involved in regulating the effect of progestogen and/or oestrogen on invasion in JEG-3.[
