定向表达COX Ⅰ功能性片段减轻叠氮钠所致线粒体功能障碍

【摘要】 将人细胞色素c氧化酶I亚型(COX Ⅰ)的功能区域性片段加入线粒体导向序列,构建pEGFP-N1-COX Ⅰ真核表达载体转染SK-N-SH细胞,探讨COX Ⅰ定向表达对叠氮钠(NaN3)引起的SK-N-SH细胞线粒体功能障碍的保护作用。结果发现转染24h后外源COX Ⅰ基因在细胞内表达明显增加,并至少持续到72h。COX Ⅰ基因功能区的线粒体定向表达不仅能维持线粒体膜电势,而且还能抑制NaN3引起的细胞增殖毒性作用。结果显示COX Ⅰ功能区域性片段可通过保护线粒体功能、减少氧化应激来减轻NaN3毒性,从而发挥保护细胞的作用。

【Abstract】 To study the protective role of targeted expression of cytochrome c oxidase Ⅰ(COX Ⅰ) domain regional fragment in mitochondrial dysfunction and oxidative stress of SK-N-SH cells resulting from sodium azide(NaN3),a mitochondrial targeting sequencing was inserted into domain regional fragment of COX Ⅰ and the plasmid pEGFP-N1-COX Ⅰ was constructed and transfected into the SK-N-SH cells.The studies showed that the expression of COX Ⅰ markedly increased at the time of 24 h after transfection and persisted up to 72 h and directional expression of COX Ⅰ domain regional fragment could not only maintain the mitochondrial potential but also inhibit the toxic effect on proliferation of cells induced NaN3.These data indicate that COX Ⅰ domain regional fragment could prevent the cells from NaN3 toxicity,which may be related to the maintenance of mitochondrial function and inhibition of ROS production.
