斑马鱼PGC-1β基因部分cDNA的克隆及其表达的初步研究

【摘要】 设计斑马鱼过氧化物酶体增殖物受体γ辅助活化因子-1β(PGC-1β)引物,扩增斑马鱼PGC-1β部分cDNA序列,并进行同源性分析。同时研究饥饿对斑马鱼PGC-1β基因mRNA表达的影响。结果表明:克隆得到斑马鱼PGC-1β部分cDNA序列长度为466bp,获得GenBank序列号为EU433886。同源性分析显示,斑马鱼PGC-1β基因同人、大鼠、小鼠、狗、鸡、金鱼同源性较低,分别为59.4%、57.5%、56.6%、59.0%、53.3%和59.0%。饥饿后,斑马鱼PGC-1β mRNA表达量显著升高。研究表明,PGC-1β在斑马鱼的能量代谢过程中可能发挥重要的作用。

【Abstract】 According to the related cDNA sequences of PGC-1β found on GenBank,the primers of zebrafish PGC-1β gene was obtained,the paritial cDNA of PGC-1β was cloned and the homologous alignment was analysed.The effect of fasting on the mRNA expression of PGC-1β in zebrafish was also studied.The result showed that the length of PGC-1β gene paritial cDNA was 466 bp,and the homology were 59.4%,57.5%,56.6%,59.0%,53.3% and 59.0% respectively between that of human,rat,mouse,dog,chicken and goldfish,which indicated that PGC-1β has lower homology on nucleotide between fish and other species.After hunger,the mRNA expression of PGC-1β increased significantly.The result indicated that PGC-1β might play important role in energy metablism in zebrafish.
