脂肪酸对小鼠前体脂肪细胞分化中脂代谢基因mRNA表达的影响
Regulation of Expression of Gene Related to Lipid Metabolism in Adipocyte Differentiation of Mouse by Fatty Acid
【摘要】 采用RT-PCR技术,分别研究短链和长链脂肪酸及P38 MAPK通路抑制剂SB20358对小鼠前体脂肪细胞分化过程中脂代谢相关基因转录表达的影响及其调控机制。结果表明:短链脂肪酸可提高PPARγ2和C/EBPαmRNA表达量(P<0.05),而长链脂肪酸上调PPARγ2、C/EBPα、FAS、SREBP-1c mRNA表达量(P<0.05),但脂肪酸对TGH表达量影响不显著(P>0.05);当P38MAPK抑制剂存在时,PPARγ2、FAS和SREBP-1c mRNA表达量显著下降(P<0.05),而C/EBPα和TGH表达量变化不显著(P>0.05)。表明脂肪酸可通过P38MAPK通路促进PPARγ2增量表达从而促进小鼠前体脂肪细胞分化。
【Abstract】 To clarify the mechanism of fatty acid regulating differentiation of preadipocyte,we examined the effect of sodium acetate(SCFA),sodium stearate(LCFA),SB203580,a P38MAPK inhibitor,on expression of gene related to lipid metabolism in differentiation of adipocyte though Real time-PCR technique.SCFA increased mRNA level of PPARγ2,C/EBPα(P<0.05),and LCFA improved expression level of PPARγ2,C/EBPα,FAS,SREBP-1c(P<0.05),whereas they did not cause any changes in TGH mRNA level(P>0.05);P38 MAPK inhibitor caused PPARγ2,FAS,SREBP-1c mRNA expression level were all significantly lower than control group(P<0.05),while C/EBPαand TGH had no significant change(P>0.05).These findings suggest that Fatty acid stimulates adipocyte differentiation by increasing mRNA level of PPARγ2 at least partly though P38MAPK.
