砷、铅胁迫对小麦幼苗毒害效应及叶片下表皮扫描电镜观察

Toxic Effects of As and Pb to Wheat Seedling and Scanning Electron-microscopic Observation on Nether Epidermis of Leaves

【摘要】 以郑州9023为材料,通过小麦发芽试验,研究了As、Pb不同质量浓度胁迫下幼苗的形态、生理指标变化,并对幼苗叶片下表皮进行了扫描电镜观察。结果表明,随着As、Pb胁迫质量浓度的增加,小麦幼苗受毒害症状逐渐加剧,其根长、芽长、根质量、芽质量均呈现出逐渐下降趋势,并与胁迫质量浓度间具有显著或极显著的一元二次函数关系;小麦籽粒α-淀粉酶活性和幼苗叶片叶绿素含量逐渐下降,叶片MDA含量、脯氨酸含量则有不同程度的升高,并且这四项指标在不同胁迫质量浓度之间均有着极显著的差异;另外,小麦叶片下表皮单位面积上的表皮毛数量增多,气孔密度增加,同时气孔开度变小。

【Abstract】 Through experiments of germination with Zhengzhou-9023 as the material,morphological indexes and physiological indexes of wheat seedling with stresses of different concentration of As or Pb were studied and nether epidermis of seedling leaves was observed with scanning electron microscope(SEM).The results showed that with the increasing of concentration of As or Pb,the toxic symptom of wheat seedling was stronger gradually: length and weight of root and plumule of seedling all decreased and significant or high significant,and quadratic equations were found between these four indexes and treatment concentrations of As or Pb;the activity of α-amylase of wheat seeds and the content of chlorophyll of seedling leaves were reduced,but the content of proline and malonaldehyde(MDA) were increased,having high significant different among different treatment concentrations;meanwhile,number of epidermal hair per unit leaf area and density of stoma all increased,but stomatal aperture decreased contrarily.
