巯基乙醇还原法提取羊毛角蛋白(英文)

【摘要】 研究用巯基乙醇还原法提取羊毛角蛋白问题,探讨了巯基乙醇的浓度对羊毛溶解率的影响规律.用质谱仪测定溶解后的角蛋白的分子量.结果显示,当巯基乙醇在溶解系统中的浓度超出一定值时不会引起进一步的溶解,反应温度是影响羊毛溶解的重要因素,随着反应温度的升高,羊毛溶解率增加,但角蛋白的大分子链会过分降解.因此,在巯基乙醇还原法中,只要采用合适的反应温度,不仅能够溶解羊毛,而且角蛋白大分子链也不会过分降解.

【Abstract】 Extraction of wool keratin by mercaptoethanol-reduction method(MERM) was investigated.The effect of concentration of mercaptoethanol(CM) on dissolution ratio of wool was examined.The mean molecular weight of keratin after dissolving was measured by a Mass Spectrometer.The results show that further dissolution did not occur while the CM concentration of solvent system exceeding a certain value.The reaction temperature was an important factor that influencing wool dissolution by MERM,and the dissolution ratio of wool was increased as the reaction temperature rising,meanwhile macromolecule chains of keratin were over-decomposed.The MERM is capable of dissolving wool without over-decomposition to macromolecular chain of keratin when the appropriate temperature is applied.
