应用液态悬浮芯片技术建立HPV基因分型方法
Development of a gene diagnosis assay for genotyping human papillomavirus using multiple suspension array
【摘要】 目的应用液态悬浮芯片技术,建立一种新的人乳头瘤病毒(HPV)基因分型方法。方法应用通用引物从HPV基因组中扩增出目的片断,与13型荧光微球偶联探针杂交,通过LuminexTM100检测HPV的型别。结果检测十三型标准株质粒,对单一标准品和混合标准品均可准确分型。结论初步建立了高通量、快速、可靠、适用于临床的人乳头瘤病毒分型基因诊断方法。
【Abstract】 Aim To develop a new method for genotyping of human papillomavirus(HPV) using multiple suspension array.Methods Common conservative primers were used to amplify HPV tarqet DNA by PCR.The PCR product were hybridized with 13 probes coupling with 13 different fluorescent microspheres.Based on the property that the fluorescent microsphere is addressable,the real timedata were measured by LuminexTM100 to detect the HPV genotype.Results The results of detecting 13 standard pl-asmids respectively using this method are all correct,and the two of mixed standard plasmids are also correctly detected.Conclusion The proposed method allowed a high through-hout,rapid and reliable detection of HPV DNA genotype.It is expected to be an extremely useful tool in clinical human papillomavirus gene diagnosis.
