白头翁对体外培养阴道毛滴虫蛋白质差异表达的影响
Effect of Pulsatilla chinensis on Trichomonas vaginalis in discrepancy protein expression
【摘要】 目的:寻找阴道毛滴虫的特异蛋白质并探讨白头翁对其蛋白质差异表达的影响。方法:用固相pH梯度二维凝胶电泳分离阴道毛滴虫总蛋白,凝胶用银染显色,PDQuest7.4.0软件分析。结果:正常对照组找到蛋白质点平均(435±17)个,匹配率为92.5%;白头翁组找到蛋白质点平均(343±9)个,匹配率为94.9%。定性分析,正常组和白头翁组共有58个表达差异点,其中只在正常组中表达的点有45个,只在白头翁组表达的点有13个;定量分析,白头翁组和正常组比较,表达增加大于2倍的点有43个,降低50%以上的点有50个。结论:建立了阴道毛滴虫正常组和白头翁组的二维凝胶电泳图谱,识别了151个差异表达的蛋白质,白头翁杀灭阴道毛滴虫的过程中上述蛋白质发生了质或量的改变。
【Abstract】 Objective:To search the characteristic proteins of Trichomonas vaginali(sT.v) and explore the influence of Pulsatilla chinensis on discrepancy protein expression.Methods: The proteins of T.v were separated with immobilized pH gradient-based two-dimensional gel electrophoresis(2-DE).After silver staining,PDQuest 7.4.0 software was used to analyse 2-DE images.Results: About 435±17 and 343±9 protein spots were found in the two maps of normal group and Pulsatilla chinensis group,respectively:The match ratio was 92.5% and 94.9% respectively.Fifty-eight different proteins were observed between the normal group and Pulsatilla chinensis group with qualitative analysis,of which 45 proteins were expressed only in the normal group,13 protein was expressed only in the Pulsatilla chinensis group;43 spots were found increased that showed a two fold increase and 50 spots were found decreased that showed 50% decrease at least as comparing to normal group with quantitative analysis.Conclusion: 2-DE protein expression profiles of T.v,including normal group and Pulsatilla chinensis group are established,and 151 different proteins are found.In the process of Pulsatilla chinensis kills T.v,the qualitative or quantitative change takes place in the above-mentioned protein.
