积雪草苷对四氯化碳诱导的大鼠肝纤维化的阻断作用
The blocking effects of glucoside of centella asiatica on rat liver fibrosis induced by CCl4
【摘要】 目的研究积雪草苷对四氯化碳诱导的大鼠实验性肝纤维化的影响。方法将SD大鼠随机分为正常对照组、模型对照组和积雪草苷治疗组,在同一环境中饲养。模型对照组和积雪草苷治疗组采用20%的四氯化碳花生油按每100g体质量0.2mL的剂量腹腔注射4周,制备大鼠肝纤维化模型,正常对照组腹腔注射等量花生油。积雪草苷治疗组于四氯化碳腹腔注射的同时给予含积雪草苷2mg的生理盐水1mL灌胃,1次/d,模型对照组和正常对照组给予等量生理盐水灌胃。第4周实验结束时,取肝组织做病理学观察,取全血做血清球蛋白、白蛋白、谷丙转氨酶、谷草转氨酶和透明质酸含量检测。结果肝组织病理学检查,正常对照组无明显病理改变,积雪草苷治疗组较模型对照组的肝纤维化分级显著降低(P<0.05);积雪草苷治疗组的血清谷丙转氨酶、谷草转氨酶和透明质酸平均升高水平也较模型对照组有显著降低(P<0.01),两组血清球蛋白和白蛋白水平无显著差异。结论积雪草苷对四氯化碳诱导的大鼠肝纤维化有明显的阻断作用。
【Abstract】 Objective To investigate the effect of glucoside of centella asiatica (GCA) to break off rat liver fibrosis induced by CCl4. Methods SD rats were randomly divided into 3 groups:the normal group,the model group and the treatment group. They were bred in same background. Rats in the model and the treatment groups were injected intraperitoneally with the mixture of CCl4 and peanut oil (1∶4) in dose of 0.2 mL/100 g for 4 weeks to induce liver fibrosis. Rats in normal group only were treated with equal peanut oil. The treatment group was given 2 mg of GCA in normal saline by gastric infusion. The model group and the normal group were infused with water. All rats were killed at the end of experiment. The pathologic changes of liver tissue stained by HE were observed under microscope. The levels of serum globulin (GLB),albumin (ALB),alanine transaminase (ALT),aspartare aminotransferase (AST) and hyaluronc acid (HA) were also measured. Results The grades of liver fibrosis in the treatment group were significantly lower than those in the model group (P<0.05). The serum levels of ALT,AST and HA in the treatment group were also significantly lower than those in the model group (P<0.01). However,the levels of the GLB and ALB were not significantly different in the two groups. Conclusion The glucoside of centella asiatica is able to break off the rat liver fibrosis induced by CCl4.
