肝纤维化大鼠模型整合素αV表达的动态观察

【摘要】 目的观察整合素(integrin)αV在实验性大鼠肝纤维化过程中表达的动态变化,初步探讨integrinαV在肝纤维化发病机制中的作用。方法将30只雄性SD大鼠随机分为模型组和正常对照组,采用腹腔内注射四氯化碳(CCl4)溶液,制造大鼠肝纤维化模型。分别在第2、4、6、8周肝纤维化发展不同阶段取肝组织,采用HE染色确定模型建立情况,Masson三色染色检测胶原沉积情况,应用逆转录多聚酶链式反应(RT-PCR)技术及Western blot检测integrinαV mRNA和α-平滑肌肌动蛋白(α-SMA mRNA)及其蛋白在肝纤维化不同时期肝组织中含量的动态变化。结果①Masson三色染色显示随着肝纤维化的发展,模型组2、4、6、8周大鼠肝组织内胶原纤维逐渐增多,均显著高于正常对照组;②RT-PCR结果显示正常大鼠肝组织中有integrinαV mRNA的表达,随着造模时间延长,纤维化程度的加重,integrinαVmRNA表达逐渐增加,α-SMAmRNA的表达也有类似变化,二者呈明显正相关关系;③Western blot检测显示随造模时间的延长integrinαV及α-SMA蛋白表达均增强。结论IntegrinαV在大鼠肝纤维化的形成过程中起重要作用。

【Abstract】 Objective:To observe the change of integrin αV in experimental liver fibrosis in rats.Methods: Liver fibrosis models were induced by carbon tetrachloride(CCl4) administration intraperitoneally and liver tissue was obtained in the 2,4,6,8 weeks during the progression of hepatic fibrosis.Distribution of collagen in the liver was assessed by-MASSON’s trichrome staintri.Expressions of integrin αV and α-SMA mRNA were evaluated by RT-PCR and by Western blotting respectively.While histopathological changes of livers were evaluated both by hematoxiline and eosin staining and by Masson’s trichrome method.Results: The expressions of integrin αV and α-SMA in model group were obviously stronger than those in the control group and the two parameters had a positive correlation(r=0.762,P<0.05).Conclusion: The integrin αV plays an important role in the pathogenesis of liver fibrosis.
