低温碱性脂肪酶产生菌的筛选及产酶培养基的优化
Screening and Optimization of Fermentation Medium of Bacterium Producing Cold-Adapted and Alkaline Lipase
【摘要】 以橄榄油为唯一碳源,从渤海湾盐碱地被油污染的土样中分离筛选出1株产低温碱性脂肪酶菌株34-5,初步酶学性质研究表明,该菌所产脂肪酶的最适温度为25,℃,最适pH为9.6.该菌的16S rDNA基因序列与伯克霍尔德氏菌(Burkholderia)的同源性为99%.摇瓶实验表明,该菌株最适产酶培养基为(g/L):淀粉10,黄豆饼粉20,玉米浆20,K2HPO4 1,聚乙烯醇大豆油乳化液20.其最高酶活为6.87,U/mL.
【Abstract】 A bacterium which produces cold-adapted alkaline lipase was isolated from soil on a selective medium that con-tained olive oil as the only source of carbon.The soil was collected from bay of Bohai.The optimal temperature and pH of the lipase were 25,℃ and pH 9.6,respectively.The sequence of 16S rDNA of strain 34-5 showed 99% homology to Burkholderia.The optimal medium for lipase production were determined as follows(g/L)s:tarch 10,bean flour 20,corn syrup 20,K2HPO4 1,emulsioned bean oil 20.The alkaline lipase activity reached maximal level of 6.87 U/mL.
