雷公藤多苷对特应性皮炎患者细胞因子表达的影响
The Effect of Tripterygium Wilfordii Polyglycosidium on Expression of Cytokines in Patients with Atopic Dermatitis
【摘要】 目的:探讨雷公藤多苷对特应性皮炎(AD)患者IL-2、IL-4、IFN-γ和IL-10细胞因子表达的影响。方法:RT-PCR方法测定与不同浓度雷公藤多苷共同培养AD患者外周血单个核细胞(PBMC)IL-2、IL-4、IFN-γ和IL-10mRNA表达。结果:(1)IL-2mRNA表达,高浓度组与对照组、低浓度组、中浓度组比较差别均有统计学意义(P<0.05或P<0.01)。中浓度组与对照组比较差别有统计学意义(P<0.01)。(2)IL-4mRNA表达,对照组与高浓度组、中浓度组和低浓度组比较差别均有统计学意义(P<0.01)。(3)IFN-γmRNA表达,高浓度组与对照组、低浓度组和中浓度组,中浓度组与对照组之间差别有统计学意义(P<0.05或P<0.01)。(4)IL-10mRNA表达,高浓度组与中浓度组之间差别无统计学意义(P>0.05),其余各组差别均有统计学意义(P<0.01或P<0.05)。结论:雷公藤多苷对体外培养AD患者PBMC中IL-2、IL-4、IFN-γ、IL-10mRNA均有抑制作用,但在低浓度时对高表达的IL-4和IL-10有抑制作用,对低表达IL-2、IFN-γ抑制作用不显著。
【Abstract】 Objective:To study the effect of tripterygium wilfordii polyglycosidium(TWP) on atopic dermatitis(AD) by detecting the expression of IL-2,IL-4,IFN-γ and IL-10 mRNA.Methods:RT-RCR was used to detect the mRNA of IL-2,IL-4,IFN-γ and IL-10 in peripheral blood mononuclear cells(PBMC)of AD cultured with different concentrations of TWP.Results(:1)The expression of IL-2 was significantly different between the high concentration group and other groups(P < 0.05 or P < 0.01),and between the control group and the middle concentration group(P < 0.01)(.2)The expression of IL-4 was significantly different between the three different concentration groups(P < 0.01).(3)The expression of IFN-γmRNA was significantly different between the high concentration group and the control,the low,the middle concentration groups(P < 0.05 or P < 0.01).(4)There was no significant difference in expression of IL-10 mRNA between the high concentration group and the middle concentration group(P > 0.05),there was significant difference among the other groups(P < 0.05 or P< 0.01).Conclusion:TWP can suppress the expression of IL-2,IL-4,IFN-γ and IL-10 mRNA,but the low concentration of TWP can suppress the high-level expression of IL-4 and IL-10mRNA in vitro.
