叶酸对局灶性脑缺血大鼠血浆同型半胱氨酸的影响
Effects of Folic Acid on Plasma Homocysteine Level in Rats with Cerebral Ischemia
【摘要】 目的:探讨叶酸对局灶性脑缺血大鼠血浆同型半胱氨酸(homocysteine,Hcy)含量的影响,为研究维生素防治脑梗死提供理论依据。方法:SD雄性大鼠32只,随机分为4组,假手术(SO)组、缺血(IM)组、缺血+叶酸低剂量[IM+LFA,4mg/(kg·d)]组、缺血+叶酸高剂量[IM+HFA,12mg/(kg·d)]组。叶酸灌胃28d后制备大脑中动脉闭塞(MCAO)模型。分别于叶酸灌胃前后和缺血后用化学免疫发光法测定血清中叶酸含量,用高效液相色谱(HPLC)法测定大鼠血浆Hcy浓度。结果:叶酸灌胃前,各组大鼠各项指标的差异均无统计学意义(P>0.05);叶酸灌胃后及缺血后,IM+LFA组、IM+HFA组血清叶酸水平高于SO组和IM组(P<0.01);叶酸灌胃后,IM+LFA组、IM+HFA组血浆Hcy水平低于叶酸灌胃前和SO、IM组(P<0.01);造模后,IM+LFA组、IM+HFA组血浆Hcy水平低于灌胃前和SO组、IM组(P<0.01),IM+HFA组低于IM+LFA组(P<0.01)。结论:补充叶酸可以提高局灶性脑缺血大鼠体内血清叶酸含量,降低血浆同型半胱氨酸水平,从而起到保护作用。
【Abstract】 Objective:To explore the effects of folic acid on plasma homocysteine level in rats with focal cerebral ischemia,so as to find theoretical evidence for the therapy and the prophylaxis of cerebral infarction with vitamin.Methods:Thirty-two male SD rats were randomly divided into four groups consisting of a sham operation group(SO),a ischemia model group(IM),a ischemia model + low dose folic acid group(IM+LFA) and a ischemia model + high dose folic acid group(IM+ HFA).The focal cerebral ischemia model was induced by occlusion of middle cerebral artery after 28 days intragastric administration.Serum folic acid contents of rats were measured before and after supplementation and after ischemia by chemical immune assay.Plasma homocysteine indices of rats were measured by high performance liquid chromatography.Results:There was no significant difference in all indices in the four groups before supplementation(P > 0.05).Serum folic acid concentration were higher in IM+LFA and IM+HFA groups than those in SO and IM groups after supplementation and ischemia(P <0.01).Levels of plasma homocysteine concentration were lower in IM+LFA and IM+HFA groups than those of SO and IM groups before supplementation(P < 0.01).After ischemia the levels of homocysteine were lower in IM+LFA and IM+HFA groups than those in SO and IM groups(P < 0.01).Conclusion:Supplementation of folic acid can increase serum folic acid concentration and reduce plasma homocysteine concentration,and thus exert a protective effect.
