3-乙基-3-甲基戊二酰亚胺致癫癎大鼠促肾上腺皮质激素和皮质醇的变化
Changes in ACTH and Cortisol Levels in Rats with Epilepsy Induced by 3-ethyl-3-methyl Glutarimide
【摘要】 目的:探讨促肾上腺皮质激素(ACTH)和皮质醇与癫癎发病的关系。方法:选用SD雄性大鼠以3-乙基-3-甲基戊二酰亚胺背部皮下注射致急、慢性癫癎,同时建立生理盐水对照组(每组20只),放射免疫方法观察癫癎发作后30min和24h血ACTH和皮质醇的变化,并与对照组比较。结果:急性致组ACTH和皮质醇水平在癫癎发作后30min时均高于对照组(P<0.01),发作后24h基本恢复正常,与对照组比较差异无统计学意义(P>0.05);慢性致组在最后一次致后30min和24h血ACTH和皮质醇水平均明显低于对照组和急性致组(P<0.01)。急、慢性致血ACTH和皮质醇在30min时均明显高于24h水平(P<0.01)。结论:癫癎发作可引起血ACTH和皮质醇水平改变,推测反复慢性致可导致下丘脑-垂体-肾上腺(HPA)轴功能紊乱。
【Abstract】 Objective:To explore the relationship between epileptic seizure and the levels of adrenocorticotrophic hormone(ACTH) and cortisol.Methods:Acute and chronic epilepsy models were induced by 3-ethyl-3-methyl glutarimide subcutaneous injection in 20 male SD rats.Rats in the control group(n=20) were injected with saline.The levels of ACTH and cortisol were detected 30 min and 24 h after epileptic seizure by radioimmunological method in two groups.Results:The levels of ACTH and cortisol were higher in 30 min after epileptic seizure in acute epileptogenic group than those in control groups(P < 0.01),and they recovered to normal range within 24 h,showing no significant difference compared with control group(P > 0.05).The levels of ACTH and cortisol were lower in 30 min and 24 h after the last epileptic seizure in chronic epileptogenic group than those in control and acute epileptogenic groups(P < 0.01).The levels of ACTH and cortisol were significant higher in 30 min than those in 24 h after epileptic seizure in acute and chronic epileptogenic group(P < 0.01).Conclusion:The changes in ACTH and cortisol were caused by epileptic seizure.Repeated chronic epileptic seizures may damage the function of hypothalamic-pituitary-adrenak(HPA) axis.
