【摘要】 目的:观察护肾固精方对单侧输尿管梗阻(UUO)大鼠肾间质纤维化(RIF)的影响,并探讨其防治RIF的作用及其机制。方法:制备大鼠单侧输尿管梗阻实验性RIF模型,随机分为3组:假手术组、单侧输尿管梗阻组、护肾固精方治疗组,观察比较各组大鼠一般情况,肾脏大体形态及病理学改变,并采用免疫组化方法检测骨形态发生蛋白-7(BMP-7)和转化生长因子-β1(TGF-β1)在各组大鼠肾组织中的表达。结果:假手术组肾脏病理未见异常;中药组大鼠术后各时相点肾组织病变较UUO组同期轻(P<0.05)。UUO组14天后肾脏中BMP-7表达较少,21天时则仅有微量表达(P<0.01),护肾固精方可以提高BMP-7表达水平,与假手术组同一时相点相比有所减弱(P<0.05)。TGF-β1在假手术组中几乎无表达,在UUO组随梗阻时间延长,TGF-β1表达逐渐增加;中药组与UUO组同一时相点比较,TGF-β1阳性表达明显减弱(P<0.05)。结论:护肾固精方可以通过提高BMP-7表达水平、降低TGF-β1表达发挥其延缓肾间质纤维化进程的作用。更多还原

【Abstract】 Objective:To observe the effect of hushen gujing prescription on rats’ renal interstitial fibrosis(RIF) caused by singal-side ureter obstruction,discuss its mechanism of preventing and curing the RIF.Methods: Models with RIF were made by ligaturing rats’ left ureters near the renal pelvis. Rats were divided into shame operation group,model group and hushen gujing prescription group,gujing prescription group rats were given herbs decoction 2ml/d,the other two were given the same volume sodium chloride, meanwhile observing and comparing the general state of health,the shape of kidney and pathological changes of each group.The expressions of BMP-7 and TGF-β1 in rats’ kidney tissue were determined by immunohistochemistry method.Results: There were no pathological changes in the shame operation group,the pathological changes in hushen gujing prescription group rats were lighten more obviuosly than those of the model group on the same stage.The expression of renal BMP-7 in the model group was less on the 14th day, only little expressed on the 21st day, the expession of renal BMP-7 was obviuosly elevated in hushen gujing prescription group, which was weaker than that of shame operation group.There was almost no TGF-β1 expression in kidneys of shame operation group, the expression was increased gradually with the prolonging of ureter obstruction time, hushen gujing prescription could decrease the expression of TGF-β1 in kidneys of rats with singal-side ureter obstruction.Conclusion: Hushen gujing prescription can prevent and cure RIF by elevating BMP-7 and lowering TGF-β1 expression.更多还原
