莲房原花青素和银杏内酯联用对东莨菪碱所致小鼠记忆获得性障碍的改善作用
The Mixture of Procyanidins Extracted from the Lotus Seedpod and Ginkgolides Ameliorates Scopolamine-induced Memory Impairment in Mice
【摘要】 目的观察莲房原花青素和银杏内酯联用对小鼠学习记忆能力的影响。方法采用Morris水迷宫筛选空间学习记忆能力相近的昆明种雄性小鼠50只,随机分为5组,即正常对照组、模型组、低剂量治疗组、中剂量治疗组、高剂量治疗组,每组10只;行为学测试开始前10min腹腔注射东莨菪碱,建立小鼠记忆获得障碍模型;3个治疗组分别灌胃50mg/kg莲房原花青素+10mg/kg银杏内酯、100mg/kg莲房原花青素+20mg/kg银杏内酯、150mg/kg莲房原花青素+30mg/kg银杏内酯;其余两组灌胃生理盐水。喂养30d后以行为学实验(Morris水迷宫法、跳台法)为指标观察莲房原花青素和银杏内酯的联合作用。结果在东莨菪碱所致小鼠记忆障碍模型中,莲房原花青素和银杏内酯联用在Morris水迷宫中可缩短潜伏期,减少游泳距离;在跳台实验中可以延长平台停留期,显著减少错误次数。结论莲房原花青素和银杏内酯联用能够改善东莨菪碱所致小鼠学习记忆障碍。
【Abstract】 Objective To study the synergic effects of lotus seedpod procyanidins (LSPC) and Ginkgolides on impairment of learning and memory in young mice induced by scopolamine. Methods Fifty male Kunming mice were screened by Morris water maze test and then randomly divided into 5 groups, 10 in each: control, model, low dose (L)-(LSPC+BIL) (50 mg/kg LSPC+10 mg/kg BIL), middle dose (M)-(LSPC+BIL) (100 mg/kg LSPC+20 mg/kg BIL), high dose (H)-(LSPC+BIL) (150 mg/kg LSPC+30 mg/kg BIL). The last three groups were treated with LSPC (dissolved in normal saline) at concentrations of 50, 100, 150 mg/kg, and Ginkgolides at concentrations of 10, 20, 30 mg/kg by gastric gavage once every day for 30 days, while the other two groups were treated with normal saline via gastric gavage. The model of learning and memory impairment was established by intraperitoneal injection of scopolamine (2 mg/kg) 10 min before the beginning of the test. Thirty days later the synergic effects of LSPC and Ginkgolides on learning and memory in young mice were tested by Morris water maze and 7 days after that step-down avoidance tests can be carried out. Results In the Morris water maze test, the groups which were treated with LSPC and Ginkgolides had significantly shorter escape latencies and swimming distances than scopolamine group. In the step-down avoidance test, all three dose groups had significantly less number of errors and longer latency than scopolamine group. Conclusion The combined use of LSPC and Ginkgolides can improve the learning and memory capacities of young mice impaired by scopolamine.
