PPARs激动剂对大鼠脑缺血再灌注早期炎症反应及血脑屏障通透性的影响

【摘要】 目的:研究过氧化物酶体增殖物激活受体(PPARs)激动剂苯扎贝特对大鼠局灶性脑缺血再灌注早期炎症反应以及血脑屏障通透性的影响,探讨PPARs激动剂是否对缺血再灌注脑损伤有保护作用。方法:制作大鼠大脑中动脉阻塞再灌注模型(MCAO/R),随机分为对照组和苯扎贝特干预组,每组10只。分别检测各组脑含水量以及脑组织伊文思蓝含量,观察PPARs激动剂对大鼠脑水肿和血脑屏障破坏程度的影响;紫外分光光度法检测脑组织髓过氧化物酶(MPO)活性;免疫组化法观察白介素-1β(IL-1β)、基质金属蛋白酶-9(MMP-9)蛋白表达变化。结果:与对照组相比较,苯扎贝特治疗组大鼠脑含水量减少(P<0.05)及脑伊文思蓝含量下降(P<0.01);缺血脑组织MPO活性降低(P<0.01);炎症因子IL-1β和MMP-9蛋白表达水平下降(P<0.01)。结论:PPARs激动剂苯扎贝特能够减轻缺血再灌注早期血脑屏障损害,其保护作用与抑制炎症有关。

【Abstract】 Objective:To investigate the effect of peroxisome proliferator-activated receptor(PPARs) agonist bezafibrate on inflammation and the permeability of blood-brain barrier after early the focal cere-bral ischemia-reperfusion,and determine its protective effect in focal cerebral ischemia-reperfusion injury.Methods:Male rat underwent middle cerebral artery occlusion and reperfusion model(MCAO/R),were randomly divided into control group(n=10) and bezafibrate-treatment(6 mg/kg) group(n=10).The rat brain water content,blood-brain barrier permeability were measured;Myeloperoxidase(MPO) activity in brain tissue was measured as an index of neutrophil accumulation;The protein expression of IL-1β and MMP-9 were examined by immunohistochemistry after cerebral ischemia.Results:Compared with control group,Bezafibrate group could significantly reduce the water content of rat brain and blood-brain barrier disrup-tion;MPO activity in the bezafibrate-treated group was significantly lower;Consistently,protein expression of IL-1β and MMP-9 in bezafibrate-treated group were also significantly down regulated than those in control group.Conclusion:The present study suggests that PPARs agonist bezafibrate has neuroprotective role by anti-inflammatory actions.
