人肺腺癌紫杉醇耐药细胞株的建立及其特性研究

【摘要】 目的建立耐紫杉醇的人肺腺癌A549(A549/Taxol)细胞株,并初步研究其生物学特性。方法应用抗肿瘤药紫杉醇以体外浓度递增和短时作用法筛选A549细胞株。用四甲基偶氮唑盐(MTT)法检测细胞的耐药指数及对多种抗癌药物的敏感性;免疫细胞化学法检测P-糖蛋白(P170)、β-tubulin等耐药相关蛋白的表达情况。结果A549/Taxol细胞的紫杉醇半数抑制浓度(IC50)是亲代A549细胞的512.8倍,群体倍增时间是亲代细胞的1.28倍,对紫杉醇、丝裂霉素、长春新碱、诺维本的耐药指数依次为512.8、66.48、4.88和2.06;而对顺铂、健择无交叉耐药。A549/Taxol细胞高表达P170,β-tubulin表达水平高于其亲代细胞系。结论成功建立了一株耐紫杉醇的A549/Taxol,并呈多药耐药特性。

【Abstract】 Objective To establish a drug-resistant cell subline A549/Taxol of human lung adenocarcinoma cells and its characterize biological properties.Methods A paclitaxel resistant human lung adenocarcinoma cell line A549/Taxol was screened by continuously exposing human lung adenocarcinoma cell line A549 to gradually increasing doses of paclitaxel.The chemosensitivities of A549/Taxol and A549 to different antineoplastic were tested and IC50 measured by MTT.Several multidrug resistant proteins including P-gp(P170) and β-tubulinand were detected by immunocytochemistry.Results Index numbers of drug resistance(IR)of A549/Taxol were 512.8,66.48,4.88 and 2.66 to paclitaxel,MMC,VCR and NVB.The cell doubling time of A549/Taxol cells were 1.28 times of A549 cells.No cross-resistance was detected to DDP and gemzer.P170 and β-tubulin were highly more expressed in A549/Taxol cell lines than A549 cell lines.Conclusion The A549/Taxol cell line shows high resistance to Taxol,and had the characteristic of multidrug resistance.
