光谱法研究头孢曲松钠与DNA的相互作用
Study on the Interaction between Ceftriaxone and DNA by Molecular Spectroscopy
【摘要】 在pH7.4的Tris-HCl介质中,以溴化乙锭为荧光探针,研究了头孢曲松钠与小牛胸腺DNA之间的相互作用.采用荧光光谱、吸收光谱、Scatchard方程、圆二色谱、热变性等手段,对作用机理进行了研究,结果表明头孢曲松钠与小牛胸腺DNA之间的作用方式为混合方式,嵌入与沟槽作用是两种主要作用方式.求得25℃时的结合常数为6.3×104L·mol-1.
【Abstract】 The interactions of ceftriaxone (CTRX) and ct-DNA have been studied by fluorescence spectroscopy using ethidium bromide (EB) as a fluorescence probe in Tris-HCl buffer solution (pH 7.4).The mechanism of the interactions between CTRX and ct-DNA was also investigated by fluorescence spectrum,absorption spectrum,scatchard equation,circular dichroism spectrum,heat denaturation etc.The experimental results showed that the interaction belongs to mixed model,the intercalation and groove binding are the two major modes in this system.The average binding constants were determined as 6.3×104 L·mol-1 at 25 ℃.
