补肾活血法对糖尿病大鼠脑缺血再灌注损伤脑组织保护研究

【摘要】 目的:探讨补肾活血法对糖尿病大鼠脑缺血再灌注损伤脑组织的保护机制。方法:采用合并糖尿病大鼠脑缺血再灌注模型,通过透射电镜对大鼠海马CA1区超微结构及细胞凋亡进行观察,并采用免疫组化法观察再灌注脑组织中NF-κB的表达。结果:糖尿病大鼠脑缺血再灌2d后,海马CA1区存在细胞坏死和细胞凋亡两种细胞死亡形式,补肾活血方治疗后未见明显的细胞坏死,神经元超微结构较模型组改善,细胞凋亡发生较模型组减轻。补肾活血方组NF-κB的阳性表达明显减少(P<0.05)。结论:补肾活血法可以通过抑制CA1区神经元坏死和凋亡而发挥抗糖尿病合并缺血性脑损伤的作用。

【Abstract】 Objective:To evaluate the effects of Bushenhuoxue method(BSHXF) on altrastructure and apoptosis of hippocampal SD rat CA1 area in cerebral tissue of diabetes and cerebral ischemic reperfusion.Methods:Altrastructure and apoptosis of hippocampal SD rat CA1 area in cerebral tissue were observed on gerbil model with diabetes and cerebral ischemic reperfusion using transmission electron microscope observe expression of NF-κB.Results:On gerbil model with ischemic injury and two days reperfusion there were two forms of cell death in cerebral tissue,neuronal necrosis and apoptosis.In BSHXF recipe group,there was no neuronal necrosis,neuronal ultrastructure was more improved than model group,neuronal apoptosis was slighter than that of model group.Conclusion:BSHXF may play the role of antagonizing diabetes and cerebral ischemia by an-tagonizing CA1 area neuronal necrosis and apoptosis.
