金银花对发热新西兰兔解热作用机制的研究

【摘要】 目的探讨金银花解热作用的中枢机制,为其在临床应用提供实验依据。方法用白细胞介素-1β(IL-1β)复制新西兰兔发热模型,在观察金银花解热作用的基础上,应用免疫组化技术检测静脉注射金银花对IL-1β作用下新西兰兔视前区-下丘脑前部(preoptic anterior hypothalamus,POAH)组织中前列腺素受体EP3(E-type prostaglandin receptorEP3)表达的影响。结果金银花静脉注射后对正常新西兰兔结肠温度没有明显影响,与生理盐水组相比,最大体温上升高度(△T)及1 h体温反应指数(TRI1)两组间无统计学意义(P>0.05);IL-1β组在静脉注射IL-1β后,结肠温度逐渐升高,△T及TRI1与生理盐水组比较,有统计学意义(P<0.01);而金银花+IL-1β组结肠温度上升峰值明显低于IL-1β组,两组△T及TRI1有显著性差异(P<0.05)。新西兰兔POAHEP3在生理状态下仅有少量表达;静脉注射IL-1β诱导实验动物发热时,EP3表达明显增强(P<0.01);而金银花能减少IL-1β引起的EP3表达(P<0.05)。结论金银花对IL-1β性发热新西兰兔的解热作用机制可能与其抑制视前区-下丘脑前部EP3的表达有关。

【Abstract】 ObjectiveTo study the antipyretic effect and mechanism of Jinyinhua injection(JYH).MethodsThe IL-1β induced febrile New Zealand rabbits were chosen as experimental models and the antipyretic effect of JYH was observed.The expression of EP3 was investigated by immunohistochemistry(IHC) in POAH.ResultsThe colonic temperature went up gradually after intravenous(i.v) IL-1β.The peak value of temperature(△T) and thermal response index(TRI1) in IL-1β treated group were higher than those in control group(P<0.01).The △T and TRI1 of in JYH and IL-1β treated group were lower than those in IL-1β treated group(P<0.05).The expression of EP3 in the POAH of IL-1β treated group was increased markedly as compared with control group(P<0.01).The expression of EP3 treated by IL-1β+ JYH in the POAH was decreased as strikingly compared with IL-1β group(P<0.05).ConclusionThe antipyretic mechanism of Jinyinhua may be related with inhibition of EP3 expression in POAH of rabbits.
