降脂宁肝胶囊对D-氨基半乳糖致小鼠急性肝损伤的保护作用

【摘要】 目的观察降脂宁肝胶囊对D-氨基半乳糖(D-Gal)致小鼠急性肝损伤的保护作用。方法将60只NIH小鼠随机分为6组:模型组,正常对照组,联苯双酯阳性对照组,降脂宁肝胶囊高、中、低剂量组。用药组小鼠分别灌胃给药,而正常对照组和模型组灌服等体积生理盐水20 ml/(kg.d),连续6 d。于末次药1 h后,除正常对照组外,其余各组小鼠均腹腔注射10%D-Gal生理盐水溶液(1000 mg/kg),24 h后采血和取肝组织,检测血清ALT、AST及肝脏MDA水平,并观察其肝组织病理变化。结果降脂宁肝胶囊高、中剂量组与模型组比较,均能显著降低血清ALT、AST及肝脏MDA水平(P<0.05,P<0.01或P<0.001),明显地改善肝脏的病理变化,并呈明显的剂量-效应关系。结论降脂宁肝胶囊对急性肝损伤模型小鼠有显著的护肝作用。

【Abstract】 Objective To observe the effect of Jiangzhi Ninggan capsule on acute hepatic injury induced by D-galactosamine(D-Gal) in mice.Methods Sixty mice were divided into six groups at random: model group,control group,Bifendate group(positive control group),Jiangzhi Ninggan Capsule groups(high dose,middle dose,low dose).The mice in treatment groups were administered medicine respectively via gavage,while the others in control and model groups were treated with the same dose of nomal saline.After six days,all of the mice except those in the control group were treated with intraperitoneal injection of 10% D-Gal(1 000 mg/kg).twenty-four hour later,the mice were killed and the blood sample and liver tissue were taken.Liver pathological changes were observed and the activities of serum ALT,AST,and the content of liver MDA were detected.Results Compared with the model group,the effect on levels of ALT,AST in blood and the content of MDA in liver tissue of Jiangzhi Ninggan Capsule groups(high dose、middle dose) were reduced significantly(respectively,P<0.05;P<0.01;P<0.001),and the liver pathological changes were lightened obviously in dose.Conclusion Jiangzhi Ninggan capsule has protective acute hepatic injury induced by D-galactosamine.
