牛磺酸对博莱霉素肺纤维化大鼠的干预作用及对TGF-β1、α-SMA表达的影响

【摘要】 目的:研究牛磺酸对博莱霉素肺纤维化大鼠的干预作用及其可能的机制。方法:54只清洁SD大鼠随机分为牛磺酸组(T组)、模型组(B组)、正常组(CN组)。T组和B组实验当天气管内滴入博莱霉素5mg/kg,CN组气管内滴入等量生理盐水。在气管内滴药后第7、14、28天每组各处死6只动物,行HE、Masson染色并做肺泡炎、肺纤维化评分,测羟脯氨酸含量、免疫组化法测转化生长因子-β1(TGF-β1)、α-平滑肌肌动蛋白(α-SMA)表达水平表达。结果:B组TGF-β1、α-SMA蛋白表达明显高于CN组和T组。B组各时间点肺泡炎评分均高于同一时间点CN组和T组,实验第14、28天肺纤维化程度及羟脯氨酸含量也高于同期CN组及T组。结论:牛磺酸可减轻博莱霉素肺纤维化大鼠的肺纤维化程度,其机制可能与抑制肺组织TGF-β1表达及成纤维细胞转化为肌成纤维细胞有关。

【Abstract】 Objective To study the effect of taurine on rats with bleomycin-induced pulmonary fibrosis and its possible mechanisms. Methods 54 Spragne-Dauley rats were randomly assigned to receive intratracheal instillation of bleomycin (group T and group B, n=18 each) or saline (group CN, n=18). 6 rats in each group were sacrificed at day 7, 14, and 28 after intratracheal instillation. Hydroxyproline concentration was detected, histological changes of lung were evaluated by HE stain and Masson’s trichrome stain, and expressions of transforming growth factor-beta1 (TGF-β1) and alpha-smooth muscle actin (α-SMA) were assessed by immunohistochemistry. Results As compared with those in group T and group CN, expressions of TGF-β1 and α-SMA were higher, the scores of alveolitis were higher at different times, and pulmonary fibrosis marks and hydroxyproline concentrations were higher at day 14 and 28 in group B. Conclusion Taurine can alleviate bleomycin-induced pulmonary fibrosis in rats, which is probably through inhibiting the expressions of TGF-β1 and the transformation of fibroblasts into myofibroblasts in lung tissue.
