白藜芦醇诱导HepG2细胞凋亡中线粒体形态功能改变

【摘要】 目的:研究白藜芦醇诱导HepG2细胞凋亡的机制,探讨白藜芦醇诱导HepG2细胞凋亡过程中线粒体形态功能的改变。方法:采用AnnexinV-FITC/PI标记结合流式细胞仪测定白藜芦醇诱导HepG2细胞凋亡的作用;电子显微镜观察白藜芦醇处理8h、12h和24h后线粒体的超微结构变化;rhodamine123标记结合流式细胞仪观察白藜芦醇处理后HepG2细胞线粒体跨膜电位(ΔΨm)的变化。结果:流式细胞术分析显示,白藜芦醇能显著诱导HepG2细胞凋亡,并呈浓度依赖性。白藜芦醇处理细胞12h后,电子显微镜检测到线粒体外膜肿胀或破裂,并可伴有线粒体嵴数量减少或消失,24h后观察到明显的细胞线粒体消失、染色质浓缩、细胞核裂解为碎块等细胞凋亡特征。流式细胞仪结果显示,经白藜芦醇处理后,rhodamine123的荧光强度明显降低,说明白藜芦醇可使ΔΨm去极化。结论:线粒体的形态和结构变化在白藜芦醇诱导HepG2细胞凋亡过程中起着重要作用,白藜芦醇诱导细胞凋亡的作用可能与其破坏ΔΨm和影响线粒体结构有关。

【Abstract】 Objective To investigate the morphological and functional changes of mitochondria in resveratrol-induced apoptosis of HepG2 cells.Methods Resveratrol-induced apoptosis of HepG2 cells was investigated by AnnexinV-FITC staining and flow cytometry.After being treated with resveratrol for 8 h, 12 h, and 24 h, morphological changes of mitochondria were observed by electronmicroscope.Transmembrane potential of mitochondria was measured by rhodamine123 combined with flow cytometry.Results Resveratrol could dose-dependently induce apoptosis of HepG2 cells.After being treated with resveratrol for 12 h, mitochondria began to swell with outer membrane broken down;for 24 h, typical apoptotic changes as mitochondria vanish, chromatin condensation, and nuclear fragmentation were observed.Resveratrol could decrease the fluorescence intensities of rhodamine123, which indicated that it could depolarize mitochondrial membranes.Conclusion Resveratrol-induced apoptosis in HepG2 cells may be involved in the collapse of mitochondrial transmembrane potential and mitochondrial morphological changes.
